possible reason is lack <strong>of</strong> vacuum at sealing <strong>of</strong> vials. Some workers have proposed that cells stand<br />
the stress <strong>of</strong> desiccation better if stored under vacuum (Puhlev et al., 2001). Although Marcotty et<br />
al. (2003) sealed the vials at atmospheric pressure in their successful attempt, it can still be<br />
speculated that vacuum sealing could improved recovery <strong>of</strong> viable <strong>sporozoites</strong>.<br />
From the negative results <strong>of</strong> the in <strong>vitro</strong> and in vivo infectivity assessment, it is evident that the<br />
product <strong>for</strong>mulations and protocols explored are unsuitable and need further work. It is<br />
recommended to investigate combinations <strong>of</strong> lyoprotectants and modifications <strong>of</strong> freeze-drying<br />
protocols keeping in mind the cost-benefit ratios <strong>of</strong> such developments. <strong>In</strong> conclusion, the<br />
desiccation tolerance <strong>of</strong> the T. <strong>parva</strong> sporozoite cell seems to be extremely low. The cell membrane<br />
is however preserved and this observation, together with a previous successful event, justifies<br />
further work in this field.
