APPENDIX V<br />

CFSE staining<br />

Isolate PBMC and count (Appendix I)<br />

Spin down at 10,000 rpm <strong>for</strong> 5 minutes<br />

Resuspend in 1 ml ice-cold PBS and transfer to eppendorf tube<br />

Add CFSE at 0.56µl/1x10 7 PBMC to the suspension.<br />

Vortex gently<br />

<strong>In</strong>cubate at 37°C in water bath <strong>for</strong>10 min in the dark<br />

Centrifuge at 4°C (10,000 rpm <strong>for</strong> 5 min)<br />

Resuspend in cold RPMI (15% FCS)<br />

Wash 3 times to remove excess stain and any DMSO (10,000 rpm <strong>for</strong> 5 min)<br />

Resuspend in culture medium if required<br />

APPENDIX VI<br />

PCR reading <strong>of</strong> in <strong>vitro</strong> titration plates<br />

Prepare the titration plates and incubate <strong>for</strong> 10 days as described in Appendix I (B)<br />

Starting with 50 µl <strong>of</strong> cell culture material in each well, add 50 µl <strong>of</strong> 0.7M ammonium<br />

hydroxide per well<br />

Suspend plates over boiling water with covers <strong>of</strong>f <strong>for</strong> 20 min<br />

Leave at room temperature <strong>for</strong> about 20 min to vaporise the Sodium hydroxide<br />

Store in refrigerator overnight<br />

Dilute extract 100 times (10 times twice) within 96-well plate (10µl extract + 90µl milli-Q ®<br />

water) with 10 minutes <strong>of</strong> shaking (ELISA shaker) at each step.<br />

Take 10 µl from each well and add to 40 µl <strong>of</strong> Master mix<br />

Do PCR with T. <strong>parva</strong> Cox-primers (Cox F/R, 40 cycles)
