upward shift (cells becoming more granular) <strong>of</strong> a cluster <strong>of</strong> cells which could be indicative <strong>of</strong><br />

apoptotic PBMC (Ormerod, 1994) in the population kept at 37°C. Figure 21 is a diagrammatic<br />

presentation <strong>of</strong> typical regions <strong>of</strong> PBMC subsets. It was further noted that the 37°C sample gave a<br />

lower mean fluorescence: 134 compared to 331.7 CFSE log units <strong>of</strong> the sample stored at 4°C.<br />

8.3.2. Exclusion <strong>of</strong> Con-A<br />

Examination <strong>of</strong> smears from the two procedures revealed a clearer presentation <strong>of</strong> cells when<br />

Con-A is excluded (Figure 22).<br />
