NARAYAN REDDY Department of Agricultural Microbiology and Bioenergy College of Agriculture, Rajendranagar, Hyderabad- 500 030 Date of Receipt : 23.11.2012 Date of Acceptance :06.02.2013 ABSTRACT Ten bacterial isolates were obtained from fifteen rhizosphere soils of Groundnut and Redgram crops in the Rangareddy district. These isolates were identified as Rhizobium, based on their cultural, morphological and biochemical characteristics and were further evaluated for Plant growth promoting attributes like phosphate solubilization, production of Indole Acetic Acid (IAA), Hydrogen Cyanide (HCN) and siderophore. All the isolates showed positive results for IAA and HCN production, while 70% of the isolates produced siderophores. Further, these isolates were subjected to in vitro antagonism against three common soil borne fungal pathogens viz., Rhizoctonia solani, Sclerotium rolfsii and Fusarium solani. Due to the production of HCN and siderophore, these isolates inhibited the mycelial growth of Rhizoctonia solani, Sclerotium rolfsii and Fusarium solani in the range of 32.30- 50.73%. Among the soil borne fungal diseases of groundnut, Web blight, caused by Rhizoctonia solani is the most common disease (Dubey, 2000). Rhizoctonia solani is a wide spread and an ecologically diverse soil-borne fungus, causing different types of diseases in many plant species. It causes root rot, stem rot, fruit and seed decay, damping-off, foliar blight, stem canker and crown rot in various crops (Guleria et al. 2007). Biological control of plant diseases using antagonistic bacteria may be considered as a promising alternative to the use of some hazardous chemical fungicides. In the present study, ten Rhizobium isolates were obtained from the rhizosphere of groundnut and redgram crops and were characterized, screened for IAA, Phosphate solubilization, HCN, siderophore production and finally for antagonistic activity with Rhizoctonia solani, Sclerotium rolfsii and Fusarium solani under in vitro conditions. MATERIALS AND METHODS Isolation of bacterial isolates Bacterial isolates were obtained from the rhizosphere of groundnut and redgram crop plants using Yeast Extract Mannitol Agar (YEMA) as described by the Vincent, (1970). Screening of pure Rhizobium isolates for Phosphate solubilization, production of indole acetic acid, hydrogen cyanide and siderophores Phosphate solubilization This test was performed following spot inoculation on Pikovskaya’s medium. Clear zone around the colonies indicates phosphate solubilization. IAA production Production of indole acetic acid (IAA) was detected as described by Brick et al., (1991). Bacterial isolates were grown for 72 h in yeast extract broth tubes at 36±2 0 C. Fully grown cultures were centrifuged at 3000rpm for 30min. the supernatant (2ml) was mixed with two drops of Orthophosphoric acid and 4ml of the Salkowaski reagent (50ml, 35% of Per chloric acid, 1ml of 0.5M Fecl 3 solution). Development of pink color indicates IAA production. HCN production HCN production was tested by the method of Lorck (1948). Yeast extract mannitol broth was amended with 4.4g glycine/lit and bacteria were streaked on modified agar plate. A Whatman filter paper no.1 soaked in 2% sodium carbonate in 0.5% picric acid solution was placed at the top of the plate. Plates were sealed with Para film and incubated at 36±2 0 C for 4 days.
