MASS SPECTROMETRY UNITHead <strong>of</strong> Unit: Bruno Manadas | Post‐Doc, PhD in Cellular Biology (<strong>2008</strong>) at University <strong>of</strong> CoimbraHead <strong>of</strong> Facility since <strong>2008</strong>Staff: Vera Mendes (technician)The Mass Spectrometry Unit is specialized inidentification <strong>and</strong> quantification <strong>of</strong> proteins fromsimple <strong>and</strong> complex samples; identification <strong>and</strong>quantification <strong>of</strong> post‐translational modifications,<strong>and</strong> identification <strong>and</strong> quantification <strong>of</strong>metabolites. The Unit is also involved in theidentification <strong>of</strong> biomarkers through proteomics<strong>and</strong> metabolomics techniques with the purpose <strong>of</strong>developing new prognosis <strong>and</strong> diagnosis methods,in collaboration with other R&D units at <strong>CNC</strong>,Biocant, <strong>and</strong> external partners.Presently, the Mass Spectrometry Unit is equippedwith state <strong>of</strong> the art technology, namely: a 4000QTRAP mass spectrometer (AppliedBiosystems/MDS Sciex), hybrid triplequadrupole/ion‐trap mass spectrometer withcapacity <strong>of</strong> MS 3 , a two‐dimensional liquidchromatography system Ultimate 3000(Dionex/LCPackings), a ExQuest (Bio‐Rad) – imageacquisition <strong>and</strong> spot picking robot <strong>and</strong> a dataprocessing station (connected to two dataacquisition stations). The unit also contains severals<strong>of</strong>tware packages <strong>for</strong> data processing, includingPDQuest <strong>and</strong> ProteomeWeaver <strong>for</strong> 2D gel analysis,Protein Pilot <strong>and</strong> PEAKS <strong>for</strong> protein identification,post‐translational modifications <strong>and</strong> de novosequencing.By combining the high resolving power <strong>of</strong> the LCsystem with the structure elucidation from themass spectrometer, the Mass Spectrometry Unit isable to identify peptides, metabolites, drugs,pesticides, among others, from complex mixtures.The Unit integrates the National MassSpectrometry Network (RNEM).4000 QTRAP mass spectrometerBidimensional chromatography modular system coupled to the4000 QTRAP spectrometer128
