Figure 6: Doma<strong>in</strong> structure <strong>of</strong> NUB1 <strong>and</strong> NUB1L, some deletion mutants, <strong>and</strong> <strong>the</strong>ir <strong>in</strong>teraction<br />

with GST <strong>and</strong> GST-<strong>FAT10</strong>. (A) The positions <strong>of</strong> <strong>the</strong> doma<strong>in</strong>s <strong>of</strong> NUB1L are <strong>in</strong>dicated<br />

by <strong>the</strong> number <strong>of</strong> <strong>the</strong> am<strong>in</strong>o acids (aa). The <strong>ubiquit<strong>in</strong></strong>-<strong>like</strong> (UBL) doma<strong>in</strong>s are<br />

shown <strong>in</strong> grey, <strong>the</strong> <strong>ubiquit<strong>in</strong></strong>-associated (UBA) doma<strong>in</strong>s are shown <strong>in</strong> white. (B) Autoradiography<br />

<strong>of</strong> <strong>in</strong> vitro transcribed <strong>and</strong> translated NUB1L mutants after separation by<br />

SDS-PAGE. A 10% aliquot <strong>of</strong> each reaction was loaded to demonstrate <strong>the</strong> amount<br />

available for <strong>the</strong> GST pull-down. The positions <strong>of</strong> molecular weight markers (<strong>in</strong> kD) are<br />

<strong>in</strong>dicated on <strong>the</strong> right. (C) Autoradiography <strong>of</strong> <strong>the</strong> NUB1L mutants shown <strong>in</strong> A after<br />

<strong>in</strong>cubation with GST coupled GSH-sepharose, wash<strong>in</strong>g <strong>and</strong> separation by SDS-PAGE.<br />

The lane occupation is <strong>the</strong> same as <strong>in</strong> B. (D) Autoradiography <strong>of</strong> <strong>the</strong> NUB1L mutants<br />

shown <strong>in</strong> A after <strong>in</strong>cubation with GST-<strong>FAT10</strong> coupled GSH-sepharose, wash<strong>in</strong>g <strong>and</strong><br />

separation by SDS-PAGE. Lane occupation is <strong>the</strong> same as <strong>in</strong> B. Only wild-type NUB1L,<br />

NUB1, <strong>and</strong> NUB1L∆UBL <strong>in</strong>teract strongly, NUB1L∆UBA3 <strong>in</strong>teracts weakly with GST-<strong>FAT10</strong>.<br />

The experiments were repeated three times with similar outcomes.<br />
