Chapter 1<br />
Figure 9: NUB1 <strong>in</strong>teracts with <strong>FAT10</strong> <strong>and</strong> accelerates its <strong>degradation</strong>. (A) HEK293 T cells<br />
were transiently transfected with HA-<strong>FAT10</strong>. After pulse labell<strong>in</strong>g with [ 35 S]-methion<strong>in</strong>e,<br />
aliquots <strong>of</strong> <strong>the</strong> cells were harvested at <strong>the</strong> <strong>in</strong>dicated time po<strong>in</strong>ts <strong>of</strong> chase, lysed <strong>and</strong><br />
anti-HA immunoprecipitations were performed. Bound prote<strong>in</strong>s were analyzed by<br />
SDS-PAGE <strong>and</strong> autoradiography. (B) Same experiment as <strong>in</strong> A, but NUB1 was coexpressed.<br />
The position <strong>of</strong> NUB1 is <strong>in</strong>dicated by an arrowhead. Molecular weight<br />
markers (<strong>in</strong> kD) are shown at <strong>the</strong> right. (C) Co-immunoprecipitation <strong>of</strong> HA-NUB1 <strong>and</strong><br />
His-<strong>FAT10</strong>. Empty vector (mock), or His-Fat10 or HA-tagged NUB1 were transfected ei<strong>the</strong>r<br />
alone or toge<strong>the</strong>r with His-<strong>FAT10</strong> (His-<strong>FAT10</strong> + HA-Nub1) <strong>in</strong>to HEK293T cells. As a<br />
positive control we used His-<strong>FAT10</strong> <strong>and</strong> HA-NUB1L. The cells were lysed <strong>and</strong> an anti-HA<br />
immunoprecipitation was performed. The prote<strong>in</strong>s bound by <strong>the</strong> antibody were separated<br />
by SDS-PAGE <strong>and</strong> analyzed by autoradiography. An arrowhead on <strong>the</strong> right<br />
side <strong>of</strong> <strong>the</strong> gel denotes <strong>the</strong> position <strong>of</strong> co-immunoprecipitated <strong>FAT10</strong>. (D) The supernatants<br />
<strong>of</strong> C were immunoprecipitated with anti His 6-antibodies to show <strong>the</strong> amount<br />
<strong>of</strong> <strong>FAT10</strong> available, <strong>the</strong> position <strong>of</strong> which is <strong>in</strong>dicated by an arrowhead. (E) Graphic<br />
evaluation <strong>of</strong> <strong>the</strong> different <strong>degradation</strong> rates <strong>of</strong> <strong>FAT10</strong> as determ<strong>in</strong>ed <strong>in</strong> A (diamonds)<br />
<strong>and</strong> B (squares).<br />
