SuperSpinner – Disposable Membrane Aeration Combined with<br />

Microcarrier Application SuperSpinner C<br />

The SuperSpinner C is the next step into<br />

microcarrier application. This technology<br />

makes the SuperSpinner ideal for experiments<br />

with microcarriers. Various types of microcarriers<br />

can be used. Sartorius BBI Systems<br />

(BBI) has developed a new polycarbonate<br />

membrane holder with an integrated magnet<br />

and an insert for microcarriers. The aeration<br />

membrane is wound around the insert for<br />

microcarriers. The holder for the microcarriers<br />

is made of polyester, the mesh size is 100 µm.<br />

The insert can be filled with various types of<br />

microcarriers which are applicable and commercially<br />

available.<br />

Functional principles and application<br />

The mode of operation of the SuperSpinner C<br />

is similar to the operation of the Super-<br />

Spinner S. Once the microcarriers are filled<br />

into the module, they can be inoculated with<br />

surface dependent cells. After a period of<br />

gentle mixing, cells attach to the microcarriers<br />

and grow under very smooth conditions<br />

protected in the module. The polyester mesh,<br />

which separates the cells attached to the<br />

microcarriers and the medium, is 100% permeable<br />

for nutrients. Cell derived products,<br />

free-suspended cells, oxygen and carbon<br />

dioxide can easily pass the polyester mesh.<br />

Due to the tumbling movement of the<br />

insert, cells are sufficiently supplied with all<br />

necessary medium components and oxygen.<br />

This technology has been proven for several<br />

years in various types of fixed bed and<br />

fluid-bed bed bioreactors.<br />

Configuration of the SuperSpinner C<br />

The SuperSpinner C consists of a 1 liter glass<br />

bottle. The center port contains the insert for<br />

microcarriers, which is horizontally wound<br />

with aeration tubing. Additional ports for<br />

feed, sampling and|or harvest connections<br />

are also included. The SuperSpinner mounts<br />

on a magnetic drive platform.<br />

Mode of operation<br />

The bottom plate of the microcarrier insert<br />

is fixed by a screw and can be dismantled.<br />

The bottom plate can be removed and the<br />

microcarrier suspension can be filled into<br />

the chamber.<br />

The complete setup is sterilized in an autoclave.<br />

Once cooled, the SuperSpinner is filled<br />

with medium. If necessary, this operation<br />

may be performed outside of a laminar flow<br />

hood using one of the ports in the center<br />

of the SuperSpinner and a pair of optional<br />

sterile connectors (female connector,<br />

Part no. BB-08809410, male connector,<br />

Part no. BB-08809402).<br />

Inoculation can be conducted in the same<br />

manner using the same sterile connectors.<br />

The SuperSpinner is then placed in an incubator<br />

on a magnetic drive platform. Aeration<br />

is initiated by switching on the air pump.<br />
