CHAPTER 3: DEVELOPMENT AND EVALUATION OF ORALLY DISINTEGRATINGTABLETS (ODTS) CONTAINING IBUPROFEN GRANULES PREPARED BY HOT-MELT EXTRUSION1.0 IntroductionThe development <strong>of</strong> oral disintegrating tablets (ODTs) h<strong>as</strong> received incre<strong>as</strong>ed interestamong researchers and pharmaceutical industries over <strong>the</strong> l<strong>as</strong>t decade. ODTs are designed todisintegrate or dissolve rapidly on contact with saliva, in <strong>the</strong> absence <strong>of</strong> additional water,compared to <strong>the</strong> traditional tablet <strong>for</strong>ms. ODTs provide several advantages <strong>as</strong> <strong>the</strong>y combine <strong>the</strong>properties <strong>of</strong> both liquid and conventional tablet <strong>for</strong>mulations [1–2] . ODTs are quickly ingestedupon introduction on <strong>the</strong> tongue, thus eliminating <strong>the</strong> need to chew <strong>the</strong> tablet, swallow it intact,or take with water. Fur<strong>the</strong>rmore, administration <strong>of</strong> ODTs is favourable to paediatric and geriatricpatients or people who find swallowing difficult and <strong>for</strong> <strong>the</strong> treatment <strong>of</strong> patients wherecompliance is difficult.However, <strong>as</strong> a result <strong>of</strong> <strong>the</strong> rapid ODT disintegration, <strong>the</strong> active substance comes incontact with <strong>the</strong> t<strong>as</strong>te buds and <strong>the</strong> need <strong>for</strong> a ple<strong>as</strong>ant t<strong>as</strong>te becomes a key <strong>as</strong>pect <strong>for</strong> patientpalatability. Thus <strong>the</strong> t<strong>as</strong>te-m<strong>as</strong>king <strong>of</strong> bitter active substances is a critical hurdle to overcome<strong>for</strong> <strong>the</strong> successful development <strong>of</strong> ODT <strong>for</strong>mulations. In general, oral administration <strong>of</strong> bitteractive substances through ODT <strong>for</strong>mulations should provide an improved degree <strong>of</strong> palatability,incre<strong>as</strong>ed patient compliance and a concomitantly beneficial <strong>the</strong>rapeutic effect. In <strong>the</strong> p<strong>as</strong>t, <strong>the</strong>methods <strong>of</strong> t<strong>as</strong>te-m<strong>as</strong>king in f<strong>as</strong>t dissolving/disintegrating tablets included sweeteners andflavors. Never<strong>the</strong>less, <strong>the</strong>se additives were not a sufficient means <strong>for</strong> complete t<strong>as</strong>te-m<strong>as</strong>king.Recent advances in technology have presented viable fomrulation alternatives to t<strong>as</strong>te-m<strong>as</strong>kbitter drugs. Several approaches have been reported which involve complexation [3, 4] , freezedrying[5] , microencapsulation [6, 7] , fluidized-bed coating [8,9] and supercritical fluids [10, 11] <strong>for</strong>t<strong>as</strong>te-m<strong>as</strong>king purposes.Hot-<strong>melt</strong> <strong>extrusion</strong> (HME) is widely used in <strong>the</strong> pharmaceutical industry <strong>as</strong> a process toprepare drug delivery systems such <strong>as</strong> granules, pellets, sustained rele<strong>as</strong>e tablets and eventransmucosal/transdermal systems. Among <strong>the</strong> o<strong>the</strong>r applications HME h<strong>as</strong> been used to incre<strong>as</strong>e<strong>the</strong> bioavailability <strong>of</strong> water insoluble active substances [12 – 14] by creating molecular dispersions.There are a few studies reported where ibupr<strong>of</strong>en (IBU) extrudates were prepared by HME usingvarious polymeric matrices [15 – 18] to obtain a powdered material. However, in <strong>the</strong>se studies, IBU<strong>for</strong>mulations were developed to provide sustained rele<strong>as</strong>e pr<strong>of</strong>iles. Recently, <strong>hot</strong>-<strong>melt</strong> <strong>extrusion</strong>w<strong>as</strong> introduced <strong>as</strong> an alternative t<strong>as</strong>te m<strong>as</strong>king <strong>technique</strong> [19] where, <strong>for</strong> example, anionic activesubstances can interact with <strong>the</strong> functional groups <strong>of</strong> positively charged polymers.
