Materials and Equipment Required (Not Provided in Kit)<br />

• Precision pipettes and multichannel pipettes suitable for delivering between 25 and 200<br />

µL. Reagent volumes listed in the Test Procedure require pipette precision less than or<br />

equal to 5%.<br />

• Graduated cylinders for wash solutions<br />

• Hard plastic or adhesive plate covers and reagent-dispensing trays<br />

• Disposable sample collection dissection instruments or sample collection device<br />

• 96-well plate reader (equipped with 450-nm and 620–650-nm filters) and washer<br />

• FastPrep* (FP120A, FP220A), Precess 48*, Precess 24* or Precellys 24* instrument<br />

• Accessory pack—dilution plates, tissue-disruption tubes with beads, extended-length<br />

homogenate transfer pipette tips and adhesive plate covers (available from IDEXX)<br />

• Personal protective equipment—safety glasses, lab coats, disposable gloves, shoe<br />

covers, hairnets and face masks<br />

• 0.5–1.0 N HCl or 1.0 N H 2 SO 4 stop solution<br />

• Sodium hypochlorite (bleach), 1N NaOH and 1N HCl, deionized water<br />

• Optional—robotic sample processor with pipetting precision of ≤2.5% as measured by<br />

CV analysis (Tecan, for example)<br />

• Optional—microtiter plate shaker (e.g., IKA MTS 2/4)<br />

• Optional—plate incubator that can maintain temperature of 32–37°C and has minimal<br />

air flow<br />

• Optional—dry heat block unit for 1.5–2 mL microfuge tubes (capable of maintaining<br />

70°C temperature)<br />

• Optional—1.5–2 mL unskirted conical screw-cap microfuge tubes<br />

Tissue Sampling and Preparation<br />

A. Brain Tissue (Bovine and Small Ruminant)<br />

1. Sampling and laboratory testing must follow the<br />

regulation (EC) No 999/2001, Annex X, Chapter<br />

C, which refers in terms of collection of samples<br />

to the latest edition of the Manual of Standards for<br />

Diagnostic Tests and Vaccines of the International<br />

Office of Epizootic Diseases (OIE) stating: “The<br />

preferred sample for immunoassay should be<br />

at, or as close to the obex as possible, but no<br />

further than 1.5 cm anterior to the obex.” Collect<br />

0.30 g (±0.05 g) of nervous tissue from the left<br />

or right side of the brain stem from the level of<br />

the obex (whenever possible) with dissection<br />

instruments, and weigh the sample to ensure<br />

the correct amount. Alternatively, the IDEXX<br />

sample collection device can be used for bovine<br />

obex collection as described in the Appendix.<br />
