Interpretation of Results
Yes No No
For the assay to be valid, the negative control mean (NCx) must run with an A 450 –A REF value
less than 0.150, and the positive control mean (PCx) must have an A 450 –A REF ≥0.400.
Calculations
Calculation of negative control mean (NCx):
NCx = A1 (A 450 – A REF ) + B1 (A 450 – A REF )
2
Calculation of positive control mean (PCx):
PCx = C1 (A 450 – A REF ) + D1 (A 450 – A REF )
Calculation of the cutoff value:
Bovine cutoff = NCx + 0.120
Small-ruminant cutoff = NCx + 0.180
NOTE: See Step 11 of the Test Procedure Chart for the definition of A REF .
Results
Interpretation of sample results is based on absorbance for the sample. Samples with A 450 –A REF
values less than the cutoff value are considered negative by the IDEXX HerdChek BSE-Scrapie
Antigen Test Kit. Samples with A 450 –A REF values greater than or equal to the cutoff are classified
as initially reactive for PrP Sc , and the homogenate should be retested in duplicate on the IDEXX
HerdChek BSE-Scrapie Antigen Test Kit.
Bovine brain retesting may be done from the original tissue homogenate or from homogenate
prepared using the optional bovine heat treatment protocol, described below. If either retest
value is equal to or greater than the test cutoff, the sample is considered positive. The sample
is considered negative when both retest replicates are less than the test cutoff. Within the EU all
initially reactive bovine samples which are negative following the retest heat treatment protocol
should be reported as negative. In the member states of the European Union, samples and
corresponding tissue giving positive rapid test results should be sent to the NRL for confirmation.
Small-ruminant samples should be retested in duplicate directly from the original tissue
homogenate - Do NOT heat treat small-ruminant homogenates. If either retest value is equal
to or greater than the test cutoff, the sample is considered positive. The sample is considered
negative when both retest replicates are less than the test cutoff.
