For the assay to be valid, the negative control mean (NCx) must run with an A 450 –A REF value<br />
less than 0.150, and the positive control mean (PCx) must have an A 450 –A REF ≥0.400.<br />

Results<br />
Interpretation of sample results is based on absorbance for the sample. Samples with A 450 –A REF<br />
values less than the cutoff value are considered negative by the IDEXX HerdChek <strong>BSE</strong>-<strong>Scrapie</strong><br />
<strong>Antigen</strong> Test Kit. Samples with A 450 –A REF values greater than or equal to the cutoff are classified<br />
as initially reactive for PrP Sc , and the homogenate should be retested in duplicate on the IDEXX<br />
HerdChek <strong>BSE</strong>-<strong>Scrapie</strong> <strong>Antigen</strong> Test Kit.<br />
Bovine brain retesting may be done from the original tissue homogenate or from homogenate<br />
prepared using the optional bovine heat treatment protocol, described below. If either retest<br />
value is equal to or greater than the test cutoff, the sample is considered positive. The sample<br />
is considered negative when both retest replicates are less than the test cutoff. Within the EU all<br />
initially reactive bovine samples which are negative following the retest heat treatment protocol<br />
should be reported as negative. In the member states of the European Union, samples and<br />
corresponding tissue giving positive rapid test results should be sent to the NRL for confirmation.<br />
Small-ruminant samples should be retested in duplicate directly from the original tissue<br />
homogenate - Do NOT heat treat small-ruminant homogenates. If either retest value is equal<br />
to or greater than the test cutoff, the sample is considered positive. The sample is considered<br />
negative when both retest replicates are less than the test cutoff.<br />
