Assembly reactions containing 25 ng of linear pUC19 vector and 0.04 pmol<br />

of each fragment were performed following individu<strong>al</strong> suppliers’ recommended<br />

protocols and using the comp<strong>et</strong>ent cells provided with the kit. The tot<strong>al</strong> number<br />

of recombinant colonies was c<strong>al</strong>culated per 25 ng of linear pUC19 vector added<br />

to the assembly reaction.<br />
