instrument settings: Centre field: 3340 G. Sweep width: 230 G. Microwave<br />
frequency: 9.452 GHz. Microwave power: 47.6 mW. Modulation Amplitude: 4.76<br />
G. Modulation frequency: 86 kHz. Time constant: 40.96 ms. Conversion Time:<br />
10.24 ms. Number <strong>of</strong> scans: 24. The amount <strong>of</strong> detected nitric oxide was<br />
determined from a calibration curve generated by incubating blood samples with<br />
known concentrations <strong>of</strong> nitrite and sodium dithionite (Na2S2O4).<br />
2.2.5 Measurement <strong>of</strong> intracellular superoxide production by<br />
HPLC detection <strong>of</strong> oxyethidium<br />
HPLC measurements served as a second, independent method for<br />
superoxide detection. Superoxide was measured in aortic rings by detection <strong>of</strong><br />
oxyethidium, the fluorescent reaction product <strong>of</strong> superoxide and<br />
dihydroethidium 198 . Vessel rings were prepared as mentioned before and<br />
incubated in a 12 well plate, containing KHB and 50 µM dihydroethidium, at<br />
37°C for 15 minutes. Inhibitors (L-NIO: 100 µM, 1400W: 10 µM, N-AANG: 10<br />
µM, L-NAME: 100 µM, apocynin: 100 µM) were added and incubated at 37°C<br />
for 30 minutes, prior to the addition <strong>of</strong> DHE. Subsequently, extracellular<br />
dihydroethidium was washed <strong>of</strong>f and the rings were incubated for 1 h for<br />
intracellular accumulation <strong>of</strong> oxyethidium. The plate was covered with<br />
aluminium foil to prevent the exposure <strong>of</strong> dihydroethidium to light. Aortic rings<br />
were then homogenized in 350 µl <strong>of</strong> ice cold methanol. A 50 µl aliquot <strong>of</strong> the<br />
homogenate was stored for protein measurements. The homogenate was<br />
filtered using a syringe top filter (0.2 µm pore size) and separated by reverse<br />
phase HPLC using a C-18 column (Nucleosil 250, 4.5 mm; Sigma-Aldrich)<br />
