eight untreated HIV-1–<strong>in</strong>fected <strong>in</strong>dividuals. The method had a limit of detection<br />

of 20% to 25% for m<strong>in</strong>or sequence variants. Pattern recognition (i.e., compar<strong>in</strong>g<br />

actual sequence data with expected wild-type and mutant sequence patterns) simplified<br />

the identification of m<strong>in</strong>or sequence variants. This real-time pyrosequenc<strong>in</strong>g<br />

method was applied <strong>in</strong> a longitud<strong>in</strong>al study monitor<strong>in</strong>g the development of PI resistance<br />

<strong>in</strong> plasma samples obta<strong>in</strong>ed from four patients over a 2 1 -year period.<br />

Pyrosequenc<strong>in</strong>g identified eight primary protease <strong>in</strong>hibitor resistance mutations as<br />

well as several secondary mutations.<br />
