DESCRIPTION OF THE EXPRESSION OF PERIPHERAL<br />

CLOCK GENES IN HUMAN INFANTS<br />

8.1 Analysis <strong>of</strong> buccal swabs <strong>in</strong> <strong>in</strong>fants<br />

This chapter describes the results from the <strong>in</strong>itial analysis <strong>of</strong> peripheral gene data<br />

obta<strong>in</strong>ed from the buccal swabs collected by the participant mothers from their<br />

<strong>in</strong>fants. Six hourly swabs were taken on two consecutive days (over 48 hours) on a<br />

weekly basis throughout the period <strong>of</strong> the study. <strong>The</strong> samples were taken<br />

concurrently with sleep and deep core body temperature measurements as well as<br />

collection <strong>of</strong> ur<strong>in</strong>e for melaton<strong>in</strong> estimation and cortisol, us<strong>in</strong>g a standard monitor<strong>in</strong>g<br />

schedule. Buccal cell sampl<strong>in</strong>g was cont<strong>in</strong>ued for four weeks after and <strong>in</strong>clud<strong>in</strong>g the<br />

week a mature temperature rhythm was noted <strong>in</strong> the <strong>in</strong>fant <strong>in</strong> most cases.<br />

Of the thirty five <strong>in</strong>fants, one family had no access to refrigeration and freezer<br />

facilities with<strong>in</strong> the home sett<strong>in</strong>g, so no samples were able to be taken and safely<br />

stored. One mother opted out <strong>of</strong> buccal swab collection due to personal preference. In<br />

total the rema<strong>in</strong><strong>in</strong>g thirty three <strong>in</strong>fants had buccal swabs collected for peripheral gene<br />

estimation. Of these, one <strong>in</strong>fant had all samples excluded due to illegible labell<strong>in</strong>g.<br />

<strong>The</strong> rema<strong>in</strong><strong>in</strong>g ten <strong>in</strong>fants had <strong>in</strong>sufficient samples or <strong>in</strong>complete data sets and were<br />

therefore excluded. Samples for any week <strong>of</strong> measurement had to <strong>in</strong>clude a m<strong>in</strong>imum<br />

<strong>of</strong> 4 consecutive six hourly samples to be <strong>in</strong>cluded <strong>in</strong> the analysis.<br />

Twenty two <strong>in</strong>fants had a total <strong>of</strong> five hundred and n<strong>in</strong>ety n<strong>in</strong>e samples processed,<br />

which are reported on <strong>in</strong> this results section.<br />
