3.3. SPGRC presentation Gene Bank Management Lerotholi Qhobela SADC Plant Genetic Resources Centre (SPGRC) SPGRC was established in 1989 as a facility to conserve plant genetic resources of the region through a network of National Plant Genetic Resource Centres (NPGRCs) thereby contributing to the well being of people of the region. The facility maintains base collection of germplasm and coordinates all matters of plant genetic resources in the region, whereas, the NPGRCs maintain active collections for use in crop development and coordinates genetic resource activities at national level. The project was fully funded by the Nordic countries from 1989 to 1999 after which SADC assumed funding 10% of cost of activities in 1999 and increased its funding by 10% annually. Member states will take full charge of the project in 2009 when the Nordic countries’ funding comes to an end. The facility is managed by a Board composed of chairmen of the National Plant Genetic Resource Committees (NPGRComs). SPGRC Main Objectives ▪ Establish, over 20 years, a regional PGR centre and a network of NPGRCs. ▪ Conserve, within the region, indigenous PGR and crop genetic resources ▪ Train PGR personnel in order to build capacity in the region. Supporting On-Farm Management & Improvement of PGRFA Pilot studies were conducted in Malawi, Zambia and Zimbabwe, aimed at developing methodologies and strategies for on-farm conservation. Follow-up activities are on-going. Farmers’ groups have been identified in some countries in the region, of which, some are actively working collaboratively with NGOs in the area of on-farm conservation (Swaziland, Zambia, Malawi). Main focus is directed to seed availability instead of crop improvement. Rural resource-poor/smallscale farmers still rely on farmer-saved seed. Germplasm Storage Facilities At SPGRC, ex-situ seed conservation is in a form of cold storage in deep freezers and up-right fridges for medium and long-term storage of seeds. Medium term storage of accessions up to five or six years entails storage of seed in airtight aluminium foil packets (sealed with an aluminium foil sealer). Accession’s passport data are attached with adhesive labels to each accession packet. For long-term storage, seeds are in bottles that are sealed with a bottle-sealing machine. Unique accession and batch reference numbers are engraved on each bottle to show the label. All passport data are kept in triplicate in an Accession Register File, Base Collection File, and all the data is electronically kept in a ’Windows’ based SPGRC Documentation and Information System (SDIS). To supplement the commercial power supply, a diesel generator is connected and automatically switches on each time there is power cut-off. This avoids defrosting in freezers and maintains viability of stored seeds. Drying Facilities and Methods ▪ Seeds are dried in a walk-in-drier which has a dehumidifier and a freezing component ▪ The dehumidifier sucks in cool dry air and releases warm moist air in the atmosphere. The drier is set at 15 – 20 o C and 15 – 20% RH.
