Finally, 16 primers with high reproducibility <strong>and</strong> clear b<strong>and</strong>s werechosen for the population study.Data analysisPopulation analysisRAPD b<strong>and</strong>s were scored as present (1) or absent (0), <strong>and</strong> a (1/0) data matrixwas constructed. Frequency <strong>of</strong> alleles was calculated by the square rootmethod. Proportion <strong>of</strong> genetic diversity residing among populations (Gst) wascalculated according to Hartl <strong>and</strong> Clark (1989), <strong>and</strong> genetic diversity (He)according to Nei <strong>and</strong> Li (1979).<strong>In</strong>dividual analysisThe UPGMA cluster analysis was carried out using PHYLIP ver. 3.57c <strong>and</strong>genetic distance (D) was calculated according to Nei <strong>and</strong> Li (1979).ResultsSeventy polymorphic RAPD b<strong>and</strong>s were obtained from 13 primers. The geneticparameters calculated from the RAPD data are summarized in Table 2.Heterozygosity (H e) within population was high (0.310) in Jambi population(Sumatra) but low in both <strong>of</strong> the Kalimantan populations (0.257 <strong>and</strong> 0.247 inSungai Mahakam <strong>and</strong> Tanah Grogot populations, respectively). The values withinsubpopulations were lower than that within the Jambi population but higherthan those within the Kalimantan populations. The G STvalue was 0.152 among3 populations. The values between the two regions (Sumatra <strong>and</strong> Kalimantan),between populations in the same region (Sungai Mahakam <strong>and</strong> Tanah Grogotpopulations, Kalimantan region), <strong>and</strong> between subpopulations (Dalek <strong>and</strong>Sadarnila subpopulation in Jambi population, Sumatra) were 0.088, 0.080, <strong>and</strong>0.065 respectively.Genetic distances between individuals (d) were also calculated. Themean value within populations was 0.341; lower than the mean <strong>of</strong> betweenpopulations (0.393), <strong>and</strong> between regions the mean value was the highest (0.406),as expected. However, the mean value within subpopulations (0.355) washigher than that <strong>of</strong> between subpopulations (0.353) or within population (0.341).The UPGMA dendrogram <strong>of</strong> each individual is shown in Fig. 1. Although thedivergence was not clear, two major clusters were observed. Cluster 1 consisted<strong>of</strong> only the individuals in Kalimantan region, but cluster 2 consisted <strong>of</strong> both theKalimantan <strong>and</strong> Sumatra regions.
