The lowest-observed-adverse-effect level (LOAEL) for chol<strong>in</strong>esterase <strong>in</strong>hibition <strong>in</strong> erythrocytes<br />

<strong>in</strong> repeat-dose studies was 10 ppm (equivalent to 1 mg/kg bw per day) <strong>in</strong> a 92-day dietary study <strong>in</strong> rats.<br />

The highest NOAEL <strong>in</strong> the database was 5 ppm (equivalent to 0.5 mg/kg bw per day) <strong>in</strong> the 92-day<br />

study <strong>in</strong> rats.<br />

In a study of acute toxicity <strong>in</strong> male volunteers given ascend<strong>in</strong>g doses of diaz<strong>in</strong>on, chol<strong>in</strong>esterase<br />

activity was not <strong>in</strong>hibited <strong>in</strong> erythrocytes at 0.21 mg/kg bw, the second highest dose tested. The<br />

highest dose was ignored, as the group comprised a s<strong>in</strong>gle volunteer. Repeat-dose studies <strong>in</strong> male<br />

volunteers given diaz<strong>in</strong>on for 28–37 days showed that, while there was some <strong>in</strong>hibition of plasma<br />

chol<strong>in</strong>esterase activity at the highest tested dose of 0.03 mg/kg bw per day, no <strong>in</strong>hibition of erythrocyte<br />

chol<strong>in</strong>esterase activity was observed.<br />

<strong>Toxicological</strong> evaluation<br />

The Meet<strong>in</strong>g identified <strong>in</strong>hibition of chol<strong>in</strong>esterase activity as the most sensitive endpo<strong>in</strong>t<br />

observed after s<strong>in</strong>gle or repeated doses of diaz<strong>in</strong>on <strong>in</strong> all species. The LOAEL for reduced<br />

chol<strong>in</strong>esterase activity <strong>in</strong> erythrocytes was 1 mg/kg bw per day and the NOAEL was 0.5 mg/kg bw<br />

per day. In consider<strong>in</strong>g the NOAEL and LOAEL identified <strong>in</strong> all repeat-dose studies, the Meet<strong>in</strong>g<br />

concluded that all species appeared to be equally sensitive and the extent of chol<strong>in</strong>esterase <strong>in</strong>hibition<br />

was not dependent on duration of dos<strong>in</strong>g, once steady-state had been achieved with<strong>in</strong> 4 weeks. After<br />

consider<strong>in</strong>g all previously evaluated data and the new studies, the Meet<strong>in</strong>g established an ADI of<br />

0–0.005 mg/kg bw based on the highest NOAEL of 0.5 mg/kg bw per day for <strong>in</strong>hibition of erythrocyte<br />

chol<strong>in</strong>esterase activity <strong>in</strong> a 92-day repeat-dose study <strong>in</strong> rats, and with a safety factor of 100.<br />
