Data for historical controls: males 6.5 ± 1.7 cm and 8.7 ± 1.2 cm; females, 6.3 ± 1.2 cm and 5.7 ± 1.7 cm.<br />

The NOAEL for general toxicity was 125 mg/kg bw on the basis of transient reductions<br />

<strong>in</strong> body-weight ga<strong>in</strong>s (<strong>in</strong>clud<strong>in</strong>g body-weight losses) and feed consumption at 500 mg/kg bw<br />

and above. The NOAEL for neurotoxicity was 2000 mg/kg bw, the highest dose tested. The<br />

toxicological significance of the transient decreases <strong>in</strong> land<strong>in</strong>g foot splay observed at all doses was<br />

questionable and was considered to be <strong>in</strong>appropriate for identify<strong>in</strong>g a NOAEL for neurotoxicity<br />

(Foss, 2005a).<br />

In a short-term study of neurotoxicity conducted <strong>in</strong> compliance with the test guidel<strong>in</strong>es of the<br />

United States EPA and OECD, groups of 10 male and 10 female Crl:CD(SD)IGS BR VAF Plus rats<br />

were fed diets conta<strong>in</strong><strong>in</strong>g thiophanate-methyl (purity, 99.7%) at a concentration of 0, 100, 500 or<br />

2500 ppm for 13 weeks (91 days). Viability, cl<strong>in</strong>ical observations, body-weights, feed consumption,<br />

functional observational battery (FOB) evaluations (which <strong>in</strong>cluded detailed cl<strong>in</strong>ical observations)<br />

and motor activity evaluations were recorded. At term<strong>in</strong>ation rats were adm<strong>in</strong>istered a comb<strong>in</strong>ation of<br />

hepar<strong>in</strong> and an anaesthetic and perfused <strong>in</strong> situ with neutral buffered 10% formal<strong>in</strong>. A gross necropsy<br />

of the thoracic, abdom<strong>in</strong>al and pelvic viscera was performed. Liver and <strong>in</strong>dividual kidney weights<br />

were recorded at necropsy, and bra<strong>in</strong> weights were recorded after additional fixation of the tissue.<br />

Five rats of each sex per group were selected for neurohistological exam<strong>in</strong>ation; and the tissues from<br />

rats selected <strong>in</strong> the control group and the group receiv<strong>in</strong>g the highest dietary concentration were<br />

evaluated.<br />
