APPENDIX 6: PROCEDURES FOR WORK WITH HAZARDOUS FRAGMENTS OF<br />

DNA<br />

Note for Category B (iv), Section 4.2<br />

When working with isolated DNA molecules or amplifying DNA molecules using<br />

techniques such as the polymerase chain reaction (PCR), there are some cases where<br />

caution is warranted. Some degree of risk may exist and the extent of this is uncertain.<br />

Such cases include:<br />

1. DNA which encodes an active oncogene product or tumour suppressor gene<br />

product, particularly when this is associated with a gene promoter with high<br />

activity in human cells. DNA containing more than one active oncogene is<br />

associated with increased risk.<br />

2. DNA encoding growth factors, their receptors or other substances that might<br />

directly or indirectly alter the growth patterns of human cells.<br />

3. DNA or RNA representing complete viral genomes or fragments with the<br />

potential to regenerate live virus. Complete genomes for HIV or papilloma<br />

viruses, for example, warrant careful handling.<br />

There is some risk that such molecules could enter the cells of the operator, the<br />

principal routes of entry being through breaks in the skin. It is therefore recommended<br />

that work of this type be carried out using gloves in order to avoid skin contact. Special<br />

care shall be taken when using needles or other sharp instruments.<br />

Precautions for handling genetically manipulated viruses with the capacity to infect<br />

human cells are described in Appendix 5.<br />
