Fig. 1. Formation of a DKP thioester via an intramolecular N to S/O acyl shift reaction. In conclusion, a peptide containing a Cys-Pro-Cys (CPC) sequence can be transformed intoa peptide DKP thioester via the N-S acyl shift reaction. The DKP thioester-forming reactionrequires acidic conditions and high temperature at this stage. Current efforts are directedtoward searching the mild reaction conditions. Recombinant proteins containing the CPCsequence would be used for the production of the peptide thioesters.AcknowledgmentsThis research was supported, in part, by The Naito Foundation and Grants-in-Aid for ScientificResearch from the Ministry of Education, Culture, Sports, Science and Technology, Japan.References1.
