All images wererecorded in 40-60 minutes after the start of the incubation.Based on our results, we propose that the uneven uptake of tested CPPs into the lumen ofGMPVs depends on the balance between the general positive charge and hydrophobicity ofthe peptide, two parameters by which CPPs are divided to classes of primary, secondaryand non-amphipathic CPPs [4]. In addition, inside GPMVs the cellular RNA and DNAmight act like attractants for R 9 and pTat. As the presence of nucleic acids in GPMVs hasbeen demonstrated before [7], and while R 9 and pTat are known to reveal affinity towardsnucleic acids [8], it is feasible that the presence of these <strong>com</strong>pounds in the lumen ofGPMVs and the respective chemical gradient drive the accumulation of cationic<strong>com</strong>pounds like nucleic acid-binding dyes or highly positively charged CPPs, especiallypTat and R 9 into vesicles.AcknowledgementsThe authors would like to thank A.
