Ingrida Augspole,Tatjana Rakcejeva, Lija DukalskaCONTENT OF SUGARS, DIETARY FIBRE AND VITAMIN CIN HYBRIDS OF ‘NANTE’ CARROTS CULTIVATED IN LATVIAFigure 2. Sample-curve of carbohydrates for carrying out chromatography analysis. The content of ascorbic acid was determined bytitration with 0.05M iodine solution (Moor et al.,2005). Twenty five grams of carrots was mixedwith 100 mL of 6 g 100 g -1 solution of oxalic acidand homogenized for 60 s, and then the sample wasfiltered. 10.0 mL of the filtrate and 2.0 mL of 1g100 g -1 starch solution was titrated until the endpointwas reached (first sign of blue colour that persistsafter 30 s). The titration was repeated in triplicate foreach sample. The ascorbic acid content was calculatedusing formula:Vsample× 5000x =, (2)V × gstandardwhereV sample– iodine amount for sample titration, mL;V standard– iodine amount for vitamin C standardsolution titration, mL;g – weight of sample, g.The total dietary fiber in these samples wasdetermined according to the AOAC approved methodNo 985.29 The experiments were carried out by usingFOSS Analytical Fibertec E 1023 system providingenzymatic processing by incubation in a thermostaticshaking water bath, residue filtration was done byFiltration Module, and protein determination - byKjeldahl (Method 46-12, 1995) nitrogen equipment. The analyses were performed in three repetitions. The samples were defatted and dried with a particlesize less than 0.5 mm. Afterwards each sample wasenzymatically digested with a–amylase incubation at100 °C, as well as with protease and amyloglucosidaseincubation at 60 °C. After digestion, the total fibercontent was precipitated by adding 95 g 100 g -1 ethanol.Later the solution was filtered and fiber was collected, dried and weighed. The protein and ash content weredetermined to correct any of these substances whichmight remain in the fiber (Prosky, 1990).Data were expressed as mean ± standard deviation;for the mathematical data processing the value ofp
