The germination of grain with deionizedwater served as a control. After germination, allsprouts were dried for 48 h at +50 °C (till moisture70 g kg -1 ); then they were ground. The experimentswere perfomed in 10 replications. Viability of barleygrain was determined before grain germination. Thesprouting activity of the grain was determined aftersprouting, whereas amount of total phenols wasdetermined after the grain had dried out.Grain viabilityTo assess grain viability, the tetrazolium test (TZ)was used. It relies on the reduction of a tetrazoliumsalt (which in its oxidized state is colorless) to aninsoluble red compound (formazan), in the presenceof any dehidrogenase activity (indicative of cellviability), thus staining respiring tissue. A total of100 grains were cut along their main axis and placedin Petri dishes with the cut surface submerged in a10 g L -1 TZ solution (2,3,5-tripheniltetrazoliumchloride), and maintained at 25 °C for 3 h. Thoseembryos in which at least 30% of their surface stainedred were assumed to be viable. The experiment wasconducted in 4 replications.Grain sprouting activitySprouting activity is the number of sprouted grains,having germinated for five days. Sprouting activitytests were conducted in all samples according to thebetween-paper (BP) method of the International SeedTesting Association. Percentage of sprouted grainwas determined after 5 days. Grains were visuallyassessed according to the ISTA rules (ISTA, 2006).The experiment was conducted in 10 replications.Total phenol content (TPC)Total phenol determination started with preparationof extracts from barley grain. Barley grain was finelyground in the laboratory mill CIATRONIC KSW2669. Four grams of ground samples were extracted10 minutes in ultrasound bath (ULTRASONS,SELECTA P) with 40 mL of solvent (7/7/6 ethanol/acetone/water (v/v/v) mixture). To reach a compromisebetween alcoholic and acetone extractions, a 7/7/6ethanol/acetone/water (v/v/v) mixture was tested.After centrifugation at 3000 min -1 for 10 min usingthe centrifuge MEDITRONIC BL-C, the supernatantwas removed and the extraction was repeated oncemore. The supernatant was collected in a 50 mlvolumetric flask and refilled by solvent till mark. TheTPC of the malt extract was determined according tothe Folin-Ciocalteu spectrophotometric method withsome modifications. First, 0.25 mL of sample weretransferred to a 25.0 mL volumetric flask containing6 mL of H 2O, to which 1.25 mL of undiluted Folin-Ciocalteu reagent were subsequently added. After1 min, 3.75 ml of 20% aqueous Na 2CO 3was added,and the volume was made up to 25.0 mL with H 2O.The control sample contained all the reaction reagentsexcept the extract. After 2 h of incubation at 25 °C,the absorbance was measured at 760 nm using thespectrophotometer JENWAY 6300 (Dabiņa-Bicka,2011). Total phenols were expressed as gallic acidequivalents.
