L’utilisation du mode 32-bit évite ainsi <strong>le</strong>s débordem<strong>en</strong>ts provoqués <strong>par</strong> la multiplication de 2 pixels d’une image 8-bit. Par exemp<strong>le</strong> la va<strong>le</strong>ur 150 multipliée <strong>par</strong> 4 devi<strong>en</strong>dra 255 <strong>en</strong> mode 8-bit au lieu de 600 <strong>en</strong> mode 32-bit. Une image 16-bit non signée peut suffire. Si on utilise <strong>le</strong> masque d’un canal (R,G ou B) <strong>avec</strong> la composante de la PLU moy<strong>en</strong>ne correspondant au même de canal de cou<strong>le</strong>ur, on obti<strong>en</strong>t une image uniforme dont la va<strong>le</strong>ur pour chaque pixel est égal à la va<strong>le</strong>ur moy<strong>en</strong>ne utilisée pour calcu<strong>le</strong>r <strong>le</strong> masque. Les masques serv<strong>en</strong>t donc a corriger <strong>le</strong>s images de la non uniformité de la réponse de la camera dans chacune des trois composantes de cou<strong>le</strong>ur On décompose l’image à traiter (fibroblaste <strong>en</strong> H2AX ou Dapi) selon <strong>le</strong>s composantes R, G et B : ImageSourcei, j Cannal R,G,B On applique sur chaque composantes R, G et B, <strong>le</strong> masque correspondant ImageCorrigée i, j Canal R,G,B ImageSource i, j i, j Canal R,G,BCanal R,G,B Les 3 composantes corrigées sont fusionnées <strong>en</strong> une image RGB qui est donc l’image fina<strong>le</strong> corrigée Nous avons réalisé un plugin d’ImageJ, « PLU_RGB_Avg », qui a pour objectif d’effectuer <strong>le</strong> prétraitem<strong>en</strong>t d’images Dapi ou H2ax à <strong>par</strong>tir de PLU(s) selon <strong>le</strong> protoco<strong>le</strong> décrit précédemm<strong>en</strong>t. Au démarrage de ce plugin, il faut choisir <strong>le</strong> répertoire de travail ; c’est à dire l’emplacem<strong>en</strong>t du fichier de log et lui donner un nom (<strong>par</strong> défaut <strong>le</strong> nom du plugin). Le plugin crée alors <strong>le</strong> fichier log récapitulatif des opérations effectuées tout au long de l’exécution du plugin. Le plugin agit sur une pi<strong>le</strong> d’images de PLU sé<strong>le</strong>ctionnées <strong>par</strong> l’utilisateur dans un répertoire. Pour faciliter la vérification du plugin, nous avons crée des images réduites composées de matrice de 85x64 pixels obt<strong>en</strong>ues à <strong>par</strong>tir des images origina<strong>le</strong>s haute résolution. Cela n’affecte <strong>en</strong> ri<strong>en</strong> <strong>le</strong> processus de prétraitem<strong>en</strong>t, si ce n’est <strong>le</strong> temps de calcul et l’espace mémoire nécessaire pour traiter <strong>le</strong>s images. Le plugin a éga<strong>le</strong>m<strong>en</strong>t été vérifié sur des images complètes. Les fichiers d’essai sont donc : Image de PLU : « fibro_PLU_1.tif » et «fibro_PLU_2.tif » Image de fibroblastes H2AX : «fibro_9.tif » Page 12
