Concentrate for easy preparation of Coomassie Blue R staining solutions.<br />

Rotiphorese ® -blue is ideal for staining proteins in SDS-PAGE gels. It saves<br />

any tedious weighing of staining powder and having to prepare the mixture.<br />

Concentrate contains Coomassie Blue R, methanol and pure acetic acid.<br />

Rotiphorese ® concentrate Blue R can be diluted with 400-600 ml deionised<br />

water to obtain the following compositions:<br />

Roti ® -Black is a kit for speedy, reproducible silver staining of proteins in<br />

free and foil-supported gels.<br />

High sensitivity in combination with very low background enables detection<br />

of proteins from 1 ng/mm 2 on. 1 kit is adequate for staining 10 gels. Staining<br />

can be performed in unsupported gels without methanol. Included is an<br />

easy-to-follow instruction manual which will guarantee reliable results.<br />

Silver staining of proteins and DNA-gels<br />

Roti ® -Black silver staining kits have been optimized to provide you with the most<br />

sensitive staining possible. Adding thiosulfate during the sensibilization and<br />

generation process reduces the background and increases contrast (acc. to Blum et al.,<br />

(1987) <strong>Electrophoresis</strong> 8:93-9). Formaldehyde should preferably be fresh when<br />

applied and it reduces the silver ions (Ag + ) to so-called silver seed crystals.<br />

We recommend to:<br />

• Wash the gels carefully after each step otherwise there may be side reactions in<br />

the form of stains, spots, increased background or secondary bands.<br />

• Fix the gels carefully. Inadequate fixation impairs band formation and increases<br />

the background. Fresh and high-quality fixatives should be used and adapted to<br />

the volume and duration of gel strength.<br />
