Chapter 3<br />

The third major aim <strong>of</strong> this thesis is to clarify the capacity <strong>of</strong> <strong>alum<strong>in</strong>ium</strong> to modify the<br />

oxidant level <strong>of</strong> certa<strong>in</strong> bra<strong>in</strong> regions and to augment the striatal DAergic<br />

neurodegeneration <strong>in</strong> a 6-OHDA model <strong>of</strong> PD. The specific objectives to reach this aim<br />

are:<br />

4) To <strong>in</strong>vestigate the <strong>in</strong> vivo effects <strong>in</strong>duced by <strong>alum<strong>in</strong>ium</strong> on <strong>in</strong>dices <strong>of</strong> <strong>oxidative</strong><br />

<strong>stress</strong> by quantification <strong>of</strong> lipid peroxidation (TBARS) and the oxidant status <strong>of</strong><br />

prote<strong>in</strong>s (PCC, PTC), <strong>in</strong> the cerebellum, ventral midbra<strong>in</strong>, cortex, hippocampus, and<br />

striatum <strong>of</strong> rats.<br />

5) To assess the <strong>in</strong> vivo effects <strong>of</strong> <strong>alum<strong>in</strong>ium</strong> on the activity <strong>of</strong> certa<strong>in</strong> antioxidant<br />

enzymes, such as SOD, GPx and CAT, <strong>in</strong> the cerebellum, ventral midbra<strong>in</strong>, cortex,<br />

hippocampus, and striatum <strong>of</strong> rats.<br />

6) To measure the <strong>in</strong> vitro effects <strong>of</strong> <strong>alum<strong>in</strong>ium</strong> on the activity <strong>of</strong> some antioxidant<br />

enzymes (SOD, GPx, CAT) and the monoam<strong>in</strong>e oxidase activity (MAO-A and<br />

MAO-B).<br />

7) To study the ability <strong>of</strong> <strong>alum<strong>in</strong>ium</strong> to potentiate the capacity <strong>of</strong> 6-OHDA<br />

adm<strong>in</strong>istered <strong>in</strong> the third ventricle <strong>in</strong> an experimental model <strong>of</strong> PD to cause<br />

<strong>oxidative</strong> <strong>stress</strong> (lipid peroxidation and prote<strong>in</strong> oxidation) <strong>in</strong> ventral midbra<strong>in</strong> and<br />

striatum and to <strong>in</strong>duce neurodegeneration <strong>in</strong> striatal DAergic term<strong>in</strong>als.
