O 14 Ribotyping as a Tool for Epidemiological Typing of Salmonella<br />

Infantis in Broiler Production<br />

Abstract<br />

The study aimed to illuminate the molecular epidemiology of Salmonella Infantis that isolated<br />

from a set of broiler farms located in a particular region. In this direction poulty litter and dust<br />

collected from 6 different broiler farms and total of 176 isolates of S. Infantis were used. Isolates<br />

were treated with restriction endonuclease PvuII, to cut DNA into smaller fragments and the<br />

fragments were separated by gel electrophoresis by using Ribotyping method. Separated fragments<br />

were transferred from gel to membrane and 16S-23S rRNA operon-specific chemiluminescent<br />

substrate was subjected to hybridization with the help of labeled probe. After hybridization<br />

band profiles that were called as Ribopattern were used for epidemiological typing of isolates<br />

in comparison with a reference database. The number and relationship of bands obtained by<br />

ribotyping were analyzed with a similarity index by using Dice coefficient and unweighted pair<br />

group method with mathematical averaging (UPGMA) in GelCompar TM (Applied Maths, Ghent,<br />

Belgium) software. The results showed that ribotyping method in showed that there is a set of<br />

similar strains were isolated from litter and dust samples in the same poultry farm thus were<br />

made comments about transmission of strains between intra or inter integrates. Strains similarity<br />

was revealed that belongs to similar and different integrated. In addition, it was revealed that<br />

strains belong to farm in small-scale region. This results in the framework the methods showed<br />

that it was capabled to do epidemiological distinction of strains.<br />
