with asbestos (e.g., chrysotile) would provide<br />
a “gold st<strong>and</strong>ard” that could be used to validate<br />
<strong>the</strong> utility <strong>of</strong> long-term inhalation studies (in<br />
rats or o<strong>the</strong>r species) for predicting human risks<br />
<strong>of</strong> exposure to various types <strong>of</strong> EMPs.<br />
3.4.3 Evaluate Toxicological<br />
Mechanisms to Develop<br />
Early Biomarkers <strong>of</strong> Human<br />
Health Effects<br />
The following scheme using acellular <strong>and</strong> cellular<br />
tests can be conducted to develop a mechanistic<br />
underst<strong>and</strong>ing <strong>of</strong> fiber toxicity <strong>and</strong> to<br />
support <strong>the</strong> development <strong>of</strong> in vivo biomarkers<br />
<strong>of</strong> effect in humans. These studies must use<br />
well-characterized EMP samples as described<br />
in <strong>the</strong> tiered testing strategy presented in Section<br />
3.4. The use <strong>of</strong> size-selected fractions <strong>of</strong><br />
EMPs could provide information needed to<br />
underst<strong>and</strong> <strong>the</strong> relationships between dimension<br />
<strong>and</strong> bioactivity.<br />
Acellular assays could include measurement <strong>of</strong><br />
<strong>the</strong> generation <strong>of</strong> ROS employing electron spin<br />
resonance (ESR) or oxidant sensitive fluorescent<br />
dyes. Evaluation <strong>of</strong> <strong>the</strong> mobilization <strong>of</strong> metal ions<br />
from EMPs could indicate cytotoxic potential.<br />
The in vitro cellular tests could include <strong>the</strong> following:<br />
• generation <strong>of</strong> reactive species measured<br />
by ESR or fluorescent dyes;<br />
• generation <strong>of</strong> inflammatory, fibrogenic,<br />
<strong>and</strong> proliferative mediators, such as TNFalpha,<br />
IL-1, <strong>and</strong> TGF;<br />
• DNA damage by comet assay;<br />
• effects on cell growth regulation by measuring<br />
cell proliferation;<br />

• effects on mitosis <strong>and</strong> aneuploidy, determined<br />
by confocal fluorescent microscopy;<br />
<strong>and</strong><br />
• signal transduction pathways, such as<br />
MAPK, <strong>and</strong> phosphoinositide-3 (PI3) kinase<br />
pathways.<br />
In vivo tests would measure markers <strong>of</strong> inflammation<br />
(e.g., BAL neutrophils, inflammatory<br />
cytokines <strong>and</strong> chemokines), fibrosis (collagen,<br />
hydroxyproline), <strong>and</strong> proliferation (BrdU<br />
assay, hyperplasia) that precede pathology.<br />
Knockout mice or pathway inhibitors in rats<br />
may be used to confirm mechanistic pathways<br />
identified in vitro <strong>and</strong> develop biomarkers for<br />
disease initiation <strong>and</strong> progression. Potential<br />
biomarkers identified in in vitro <strong>and</strong> in vivo<br />
studies would be evaluated in human populations<br />
with known exposure to EMPs, <strong>and</strong> <strong>the</strong><br />
type <strong>and</strong> extent <strong>of</strong> <strong>the</strong> relationships between<br />
<strong>the</strong> marker <strong>and</strong> clinical signs <strong>of</strong> disease could<br />
be determined.<br />
3.5 Develop Information <strong>and</strong><br />
Knowledge on Occupational<br />
Exposures to <strong>Asbestos</strong><br />
<strong>Fibers</strong> <strong>and</strong> <strong>O<strong>the</strong>r</strong> EMPs <strong>and</strong><br />
Related Health Outcomes<br />
Many studies have been published concerning<br />
occupational exposures to asbestos fibers <strong>and</strong> associated<br />
health effects. These studies have formed<br />
a knowledge base that has supported increased<br />
regulation <strong>of</strong> occupational asbestos exposures<br />
<strong>and</strong> substantial reductions in asbestos use <strong>and</strong><br />
asbestos exposures in <strong>the</strong> United <strong>State</strong>s over <strong>the</strong><br />
past several decades. But, as this Roadmap makes<br />
clear, much less is known about o<strong>the</strong>r types <strong>of</strong><br />
mineral fibers <strong>and</strong> EMPs in terms <strong>of</strong> occupational<br />
exposures <strong>and</strong> potential health effects.<br />
