Durante los útlimos años hemos estudiado la función de los genes Hox Ultrabithorax (Ubx) y Abdominal-B (Abd-B) en Drosophila. Hemos determinado la expresión y función de las dos isoformas codificadas por el gen Abd-B y establecido las interacciones entre estas dos proteínas, y las de ambas con el gen Hox abdominal-A (abd-A), en el desarrollo de la genitalia. Hemos encontrado que, al contrario de lo que ocurre en el embrión, Abd-B mantiene la expresión de abd- A, lo que contradice la regla general de que los genes Hox que se expresan en regiones posteriores (como Abd-B) reprimen la expresión de los genes más anteriores (como abd-A). Hemos descubierto asimismo que Ubx controla la vía de señalización de Decapentaplegic (Dpp), un morfógeno necesario para el crecimiento y formación del patrón en distintos apéndices, en el desarrollo de los halterios (apéndices metatorácicos homólogos a las alas pero mucho más pequeños). Ubx realiza este control actuando sobre esta vía a distintos niveles. Ubx reduce tanto la producción como la movilidad del morfógeno Dpp y esto explica la reducción del tamaño del halterio con respecto a la del ala. Finalmente, hemos estudiado el desarrollo de las uniones entre los distintos segmentos de la pata. La formación de estas uniones requiere la apoptosis de algunás de sus células, producida al formarse discontinuidades bruscas en la actividad de Dpp. Esta proteína forma gradientes de actividad en cada segmento distal de las patas, con niveles de actividad mínimos en la parte más proximal de cada segmento y máximos en la distal. La confrontación de los niveles máximos de un segmento y mínimos del siguiente origina que se mueran estas células adyacentes con niveles de actividad de Dpp tan dispares y se formen las uniones correctas. The Hox genes determine the different structures along the anteroposterior axis of the animals. During the past years we have studied the function of the Ultrabithorax (Ubx) and Abdominal-B (Abd-B) Hox genes in Drosophila. We have determined the expression and function of the two isoforms encoded by Abd-B in the formation of the genitalia and established the interactions between these two proteins as well as the interactions between any of them with the Hox gene abdominal-A (abd-A). We have found that, contrary to what happens in the embryo, Abd-B maintains abd-A expression, what contradicts the rule that posteriorlyexpressed Hox genes, like Abd-B, repress those expressed more anteriorly, like abd-A. We have also studied the regulation by Ubx of the Decapentaplegic (Dpp) signaling pathway. Dpp is a morphogen that controls size and pattern of the appendages and we have found that Ubx controls the Dpp pathway at different levels in the formation of the halteres, appendages homologous to wings but much smaller. Ubx does this by regulating Dpp expression and mobility, and this control is responsible for the smaller size of the halteres with respect to that of the wings. Finally, we have studied the development of leg joints in the distal leg segments. The development of these structures requires apoptosis in some of their cells, caused by the formation of sharp boundaries of different Dpp activity. Gradients of Dpp activity are formed in each segment, with higher levels in the distal part and lower levels in the proximal one. The sharp discontinuities formed between maximal and minimal levels at the distal part of each segment are responsible for the death of adjacent cells and for the correct formation of the joints.
