y levaduras<br />
Resumen <strong>de</strong> investigación<br />
Nuestro investigación está encaminada a conocer los<br />
mecanismos moleculares <strong>de</strong> la regulación <strong>de</strong> la expresión<br />
<strong>de</strong> genes <strong>de</strong> Streptomyces y levaduras implicados en la<br />
síntesis <strong>de</strong> moléculas <strong>de</strong> interés biotecnológico. Durante los<br />
últimos dos años hemos continuado el estudio <strong>de</strong> los<br />
clusters biosintéticos <strong>de</strong> los antibióticos nucleosídicos<br />
puromicina (pur) y A201A (ata) <strong>de</strong> Streptomyces y <strong>de</strong><br />
distintas proteínas con actividad glucosidasa /<br />
glicosiltransferasa <strong>de</strong> levaduras que puedan ser empleadas<br />
en la obtención / modificación <strong>de</strong> oligosacáridos.<br />
Un 44% <strong>de</strong> la masa <strong>de</strong> A201A lo constituyen residuos<br />
glucídicos (furanosa insaturada y D-ramnosa) que sin duda<br />
están implicados en el control <strong>de</strong> las propieda<strong>de</strong>s<br />
biológicas-farmacológicas <strong>de</strong>l antibiótico. Estamos<br />
caracterizando los genes implicados en la incorporación /<br />
modificación <strong>de</strong> estos residuos. Nuestro análisis se centra<br />
en los genes ata5, ata7, ata10, ata12, ata13 ata14 y ata17;<br />
la mayor parte <strong>de</strong> estos han sido ya inactivados y los<br />
productos generados en cada caso están siendo<br />
estudiados. Se intentarán obtener nuevas moléculas con<br />
actividad antibiótica basándonos en la flexibilidad <strong>de</strong><br />
sustrato que muestran las glicosiltransferasas <strong>de</strong>scritas.<br />
El campo <strong>de</strong> los oligosacáridos prebióticos como ingredientes<br />
funcionales, en alimentación, se ha <strong>de</strong>sarrollado <strong>de</strong> manera<br />
espectacular en los últimos años. Estamos caracterizando y<br />
estudiando distintas activida<strong>de</strong>s glicosiltransferasa <strong>de</strong><br />
levaduras pertenecientes a los géneros Phaffia,<br />
Schwanniomyces y Rhodotorula que sintetizan distintos tipos<br />
<strong>de</strong> oligosacáridos con propieda<strong>de</strong>s prebióticas. Conocer la<br />
relación que existe entre la estructura, función y especificidad<br />
<strong>de</strong> estas enzimas es uno <strong>de</strong> nuestros objetivos principales.<br />
Estamos modificando posiciones concretas y aleatorias <strong>de</strong><br />
estas proteínas para aumentar/modificar su actividad<br />
transferasa, su inmovilización a soportes sólidos y favorecer<br />
su utilización industrial.<br />

Research summary<br />
Our research is focussed in knowing the molecular<br />
mechanisms regulating the expression of some<br />
Streptomyces and yeast genes, which produce novel<br />
compounds of biotechnological interest. During the last<br />
years, we are continuing to study the biosynthetic cluster of<br />
the nucleosi<strong>de</strong> antibiotics puromycin (pur) and A201A (ata)<br />
and also some yeast proteins with glucosidase/<br />
glycosyltransferase activity able to produce/modify<br />
oligosacchari<strong>de</strong>s.<br />
The monosacchari<strong>de</strong> residues constitute about 44% of the<br />
A201A mass (unsaturated furanose and D-rhamnose),<br />
which must be related with the biological-pharmacological<br />
properties of this antibiotic. We are characterizing the<br />
involved genes in the incorporation/modification of these<br />
residues. Our analysis is focused in ata5, ata7, ata10, ata12,<br />
ata13, ata14 and ata17 genes; most of these have been<br />
already inactivated and the generated products are being<br />
characterized. The biosynthesis of novel antibiotics will be<br />
attempted based on the low substrate specificity previously<br />
<strong>de</strong>scribed for glycosyltransferases.<br />
The field of prebiotic oligosacchari<strong>de</strong>s as functional<br />
ingredients in food has <strong>de</strong>veloped substantially in the last<br />
few years. We are characterizing and studying several<br />
glycosyltransferases from Phaffia, Schwanniomyces and<br />
Rhodotorula yeast genera, which produce different prebiotic<br />
oligosacchari<strong>de</strong>s. To know the structure-function-specificity<br />
relationships are ours priority objectives. We are carrying out<br />
structural modifications of theses proteins in or<strong>de</strong>r to<br />
increase/modify their transglycosylase activity, to modulate<br />
their immobilization on acrylic polymers and to improve their<br />
industrial applications.<br />
