schizofrenici, coerentemente con le evidenze di studi che avevano<br />
individuato una patologia sinaptica in queste regioni in caso di<br />
schizofrenia 19-20 . Sono state condotte anche alcune analisi esplorative<br />
rispetto alla potenziale influenza dell’abitudine al fumo.<br />
Discrepanze, invece, sono emerse rispetto ai risultati di alcuni<br />
studi post-mortem per la sinaptofisina condotti nelle stesse<br />
regioni 9 . Secondo Onwordi e colleghi, le discrepanze possono riflettere<br />
il fatto che la ricerca inglese ha misurato il legame della<br />
SV2A mentre gli studi post-mortem hanno misurato altre proteine<br />
sinaptiche. Non va poi tralasciato che nelle precedenti indagini<br />
condotte post-mortem su individui schizofrenici, anche l’età <strong>dei</strong><br />
pazienti può aver inciso sul risultato.<br />
Sicuramente un punto di forza dello studio del Medical Research<br />
Council sta nell’uso di una valutazione in vivo <strong>dei</strong> livelli di<br />
proteine sinaptiche, scelta che elimina già in premessa l’influenza<br />
esercitata dalla causa della morte. Dal punto di vista tecnico, inoltre,<br />
[11C] UCB-J è uno strumento di imaging affidabile che ha<br />
mostrato una buona ripetibilità 21 .<br />
Tutti i partecipanti sono stati sottoposti a scansione PET con<br />
[11C] UCB-J. I dati PET sono stati acquisiti per 90 minuti. L’elaborazione<br />
e la modellazione delle immagini sono state condotte<br />
utilizzando la versione 4.3.7 del software MIAKAT.<br />
<strong>Il</strong> volume di distribuzione del tracciante è stata la principale<br />
misura di esito, con la concentrazione della sostanza biochimica<br />
radioattiva nella regione target del tessuto. Le immagini PET sono<br />
state registrate sull’imaging a risonanza magnetica di ciascun soggetto,<br />
e così, corrette. Sono state generate le curve attività-tempo<br />
regionali per ciascuna area di interesse (ROI).<br />
«Dobbiamo sviluppare nuovi trattamenti per la schizofrenia.<br />
Questa proteina SV2A potrebbe essere un nuovo bersaglio di trattamenti<br />
innovativi capaci di ripristinare la funzione sinaptica», ha<br />
detto in una nota divulgativa il professor Howes.<br />
La dottoressa Ellis Onwordi ha dichiarato: «La schizofrenia<br />
è un disturbo altamente debilitante e le opzioni terapeutiche sono<br />
troppo limitate per molti pazienti. Per individuare nuovi trattamenti,<br />
in futuro abbiamo bisogno di studi come questo per far luce sulle<br />
dinamiche di un cablaggio straordinariamente complesso come<br />
quello del cervello umano che viene alterato da questa malattia. Avere<br />
scansioni che possono caratterizzare la distribuzione di circa 100<br />
trilioni di sinapsi nel cervello vivente e trovare differenze nella loro<br />
distribuzione tra persone, con e senza schizofrenia, rappresenta un<br />
progresso significativo nella nostra capacità di studiare la malattia».<br />
Secondo i ricercatori, dunque, la scoperta potrebbe aprire la strada<br />
a nuove cure e, soprattutto, far conoscere più a fondo le dinamiche<br />
di quella intricata rete di snodi che è il nostro cervello.<br />
