61Sabine Montaut with/avec Nelson Belzile & Yuwei ChenHigh Performance Liquid Chromatograph for the separationof natural productsA grant of $75,989.Une subvention de 75 989 $.Research Tools and Instruments – Subventions d’outils etd’instruments de rechercheThe High Performance Liquid Chromatograph is a key instrument thatwill allow us to analyze and separate small and large amounts of a wide varietyof natural products and mixtures of compounds. The mixture travelsthrough a column under pressure containing a stationary phase. For a givenmobile phase, the products are eluted according to their polarities. Thediode-array detector attached to the instrument visualizes the effectivenessof the separation and provides structural information. After creating our ownproduct-separation protocols, this automated instrument will acquire, integrateand store all the data. The High Performance Liquid Chromatographis essential for plant extract analysis, natural product separation and quantificationof natural products and new biologically active metabolites in plantextracts. A library of known compounds will be created so that they can beeasily identified in complex mixtures. We will use this instrument to investigate the phytochemical compositionof wild cruciferous plants that have not been screened for potential cancerchemoprotection or antioxidant properties. In fact, cancer chemopreventiveactivity of cruciferous vegetables (broccoli, cabbage, cauliflower, kale, Brusselssprouts) is believed to be due to sulfur-containing metabolites (glucosino1ates,isothiocyanates), flavonoids and other phenolic compounds. TheHigh Performance Liquid Chromatograph will also be used to screen endemicplants of Canada, plants used in folk medicine, plant families alreadyknown to possess natural cancer chemopreventive agents or very promisingnew candidates. The research is expected to give a better understanding of the relativecontributions of various components of these plants to cancer risk reduction, and provide fresh avenues for developing novel anti-cancer drugs.
