Inhibition of oxytocin in repeated milkingflow ceased again 10 IU of OT was injected intravenously to remove theremaining milk including residual milk. Milk flow was recordedcontinuously and blood samples were collected via jugular vein cannulaat 1-min intervals from 1 min before start of milking until i.v. injection ofOT. The inhibition of milk ejection and its normalization during repeatedmilking in unfamiliar surroundings was not influenced by naloxonetreatment. Therefore, any role of endogenous opioids in the inhibition ofmilk ejection in unfamiliar surroundings could not be demonstrated. Therewere not differences in cortisol release during control milking and allrelocations between naloxone and control group.In addition, the effect of exogenous ACTH 1-24(8 IU) i.v. on the release ofcortisol related to the response of cows milked in unfamiliar surroundingswas studied. Cows with totally inhibited milk ejection in response tovaginal stimulation during milking after first relocation had numericallylower cortisol levels (8.8±3.4 ng/ml; AUC/min) in response to ACTH thancows with at least partial milk ejection (38.7±12.9 ng/ml). Thus animalswith higher adrenal response to ACTH seemed to have less severeinhibition of milk ejection.ReferencesBicknell, R. This acceptation of biologicalpotentials and limitations of dairy cows allows us to milk them fast andcomplete and reduce or exclude any adverse effects on the cows. Thedevelopment of new technologies gives us a new opportunity tounderstand and include biological responses of dairy cows into the milkingprocess.On the basis of the milk flow profiles analysed at the quarter and udderlevel the possible relationships between the duration of decline phase ofmilk flow and SCC (somatic cell count) were investigated. 40 Holsteincows, free of clinical mastitis and on their first to third lactation, wereused. In total 1760 quarter and 440 udder milk flows were recorded duringsix consecutive days. At the last evening and morning milking of this periodthe milk samples from all quarters for SCC were collected. For the analysisall quarters and cows were classified in three different groups based onthe duration of the decline phase – less than 40 s, between 40 and 80 s, andmore than 80s for quarter analysis and less than 120 s, between 120 and200 s and more than 200 s for udder analysis. The statistical analysis wascarried out with the Genstat package (Genstat 5 Comittee, 1999).At the udder level no effect of the duration of decline phase on SCC wasfound. With the increase of the decline phase the milk yield alsosignificantly increased. The plateau phase increased only slightly whenthe decline phase increased.ICAR Technical Series - No 7277
