DIVERSITY OF PHYTOPATHOPGENIC BACTERIA OF THE GENUSBURKHOLDERIA IN BULGARIAY. Kizheva 1 , P. Hristova 1 , N. Bogatzevska 2 , S. Tishkov 1 , V. Chipeva 1 ,P. Moncheva 11 Biological Faculty, Sofia University “St. Kliment Ohridski”, 1164 Sofia,Bulgaria; 2 Plant Protection Institute, Kostinbrod, BulgariaThe aim was to study species and intraspecies diversity of bacteria of the genusBurkholderia isolated from onion bulbs in Bulgaria. Microbiological and molecularmethods were used for the aim realization. The strains were isolated from infectedplant material. The type cultures of Burkholderia gladioli pv gladioli, Burkholderiagladioli pv alliicola, Burkholderia cepacia as well as clinic isolates of B. cepaciawere used as controls. The identification of the pathogenic strains was performedby BIOLOG system and three species were recorded: B. cepacia, B. gladioli and B.pyrrocinia. The intraspecies diversity was studied on the basis of the metaboliteprofile of the strains, obtained by BIOLOG using cluster analysis. The strains wereseparated into seven clusters, which showed the species and intraspecies diversity,which was greater among the strains of B. gladioli. The clinic isolates of B.cepacia distinguished from the phytopathogenic ones. The RFLP-analysis wascarried out with 16S rDNA amplified fragment by the pair of universal primers (9fand 1542r) and endonucleases AluI, TasI and TaqI. The selected enzymesdiscriminated B. cepacia from B. gladioli as well as the two pathovars of B.gladioli. The enzyme AluI revealed polymorphism among the B. gladioli pvgladioli strains. This analysis did not confirmed species belonging to the strainsidentified as B. pyrrocinia by BIOLOG. This study is the first phenotypic andgenotypic characterisation of B. cepacia and B. gladioli in Bulgaria and suggests apolyphasic approach to differentiation of the two pathovars of B. gladioli andintrasepcies diversity.
