(<strong>Gamma</strong> room)” <strong>and</strong> “Carbon ion-beams” irradiation methods. These shoot<br />

tips were developed in a solid medium that consisted of MS medium<br />

(Murashige <strong>and</strong> Skoog, 1962) supplemented with BA (2.25 mg L -1 ), IAA<br />

(0.05 mg L -1 ), sucrose (20 g L -1 ) <strong>and</strong> agar (9 g L -1 ) at pH 5.6.<br />

For fast propagation, during pre- <strong>and</strong> post-irradiation, the<br />

explants were transferred to the multiplication liquid medium by dividing<br />

the corm of each in the case of “<strong>Gamma</strong> rays (<strong>Gamma</strong> room)”. The<br />

multiplication medium was same as the initiation medium but without agar.<br />

For “Carbon ion-beams” irradiation, banana explants as thin as<br />

2 mm were requested to allow total penetration of the ion beam. Prior to<br />

irradiation, from April 28 to May 10, 2005, an experiment was conducted<br />

to clarify the regeneration rate of the thinner banana explants. Cultivars<br />

‘FHIA-01’ <strong>and</strong> ‘Williams’ were used. Two types of slicing or cutting<br />

methods were applied to the banana corms shoot tips (both vertical <strong>and</strong><br />

horizontal slices). The slices were placed in liquid medium <strong>for</strong> regeneration,<br />

containing MS medium supplemented with BA (5 mg L -1 ), <strong>and</strong> sucrose (20<br />

g L -1 ), at pH 5.6. Twenty-five slices were placed into a 300 mL Erlenmeyer<br />

flask containing 100 mL of medium. Five flasks (replications) containing<br />

the explants were prepared. The explants were stirred in a shaker at 100<br />

rpm. Regeneration rate (%) <strong>and</strong> weight of explants were recorded. From the<br />

results, the highest number of regenerated plants obtained using either of<br />

the two methods were selected <strong>and</strong> applied in the establishment of the<br />

experiment <strong>for</strong> ion-beams irradiation.<br />

In both “<strong>Gamma</strong> rays (<strong>Gamma</strong> room)” <strong>and</strong> “Carbon ion-beams”<br />

irradiation methods, the multiplication medium containing the explants was<br />

stirred in a shaker at 100 rpm <strong>and</strong> in all maintenance stages, temperature<br />

was kept at 27 o C. After irradiation, initiation solid medium, multiplication<br />

liquid medium <strong>and</strong> regeneration solid medium were used to propagate the<br />

explants into a largest population to study the mutations.<br />
