tures at sites <strong>of</strong> inflammation. The group demonstrated that <strong>the</strong><br />
homeostatic chemokine receptors CXCR5 and CCR7 are critical signaling<br />
molecules in lymphoid neo-genesis during chronic inflammation.<br />
To this end, <strong>the</strong>y have established a novel mouse model <strong>of</strong> antigeninduced<br />
arthritis (AIA), which allows <strong>the</strong> efficient induction <strong>of</strong> ectopic<br />
lymphoid follicles, a hallmark <strong>of</strong> human rheumatoid arthritis. Ano<strong>the</strong>r<br />
recent accomplishment <strong>of</strong> <strong>the</strong> group was to show that CCR7 is also a<br />
key regulator <strong>of</strong> homeostatic lymphocyte recirculation through body<br />
cavities and non-lymphoid peripheral tissues.<br />
Autoreactive T cells, which have escaped <strong>the</strong> regulatory mechanisms<br />
<strong>of</strong> <strong>the</strong> immune system, may cause severe autoimmune diseases.<br />
However, <strong>the</strong>y may also be <strong>the</strong> key to a successful induction <strong>of</strong> tumorspecific<br />
immune responses since by definition, tumor-associated<br />
antigens (TAA) are self-antigens that require a break <strong>of</strong> tolerance. The<br />
group <strong>of</strong> Kirsten Falk and Olaf Rötzschke was <strong>the</strong> first to describe an<br />
effector memory-like subset <strong>of</strong> regulatory T cells, which can be identified<br />
by expression <strong>of</strong> <strong>the</strong> chemokine receptor CCR6. The positioning<br />
and phenotype <strong>of</strong> <strong>the</strong>se regulatory T REM cells suggests that <strong>the</strong>y are<br />
meant to control potentially destructive autoimmune responses in<br />
inflamed tissues. These results have been corroborated by demonstrating<br />
that MS patients have significantly reduced numbers <strong>of</strong> a specific<br />
T REM subset. Notably, this subset is characterized by <strong>the</strong> expression<br />
<strong>of</strong> an ecto-ATPase (CD39) suggesting a novel role <strong>of</strong> extracellular<br />
ATP in immune regulation.<br />
The immune system usually fails to reject established tumors. In order<br />
to develop effective cancer <strong>the</strong>rapies, it is <strong>the</strong>refore essential to<br />
understand <strong>the</strong> molecular interaction <strong>of</strong> tumor cells, immune cells,<br />
and <strong>the</strong> tumor microenvironment. The group <strong>of</strong> Thomas<br />
Blankenstein focuses on immunological mechanisms involved in <strong>the</strong><br />
activation <strong>of</strong> tumor-associated T cells and tumor rejection. They have<br />
now shown that spontaneous tumors do not escape T cell recognition<br />
but instead induce T cell tolerance, hence pointing out a major obstacle<br />
for immunosurveillance against spontaneous tumors. The groups<br />
<strong>of</strong> Wolfgang Uckert, Thomas Blankenstein and Antonio Pezzutto<br />
have a strong interest in cancer immuno<strong>the</strong>rapy based on adoptive T<br />
cell transfer. In a collaborative effort <strong>the</strong>y have developed novel<br />
methods to graft antigen specificity onto primary human T cells,<br />
<strong>the</strong>reby generating T cells with function and specificity comparable to<br />
<strong>the</strong> original T cell clone from which <strong>the</strong> T cell receptor (TCR) was isolated.<br />
Based on <strong>the</strong>se promising results, first clinical studies are<br />
planned to treat Epstein-Barr virus (EBV) associated diseases such as<br />
Hodgkin’s disease and post-transplant lymphoproliferative disorders<br />
(PTLDs).<br />
Over <strong>the</strong> past years, <strong>the</strong> group <strong>of</strong> Bernd Dörken has received wide<br />
international recognition for pioneering work on <strong>the</strong> molecular<br />
pathogenesis <strong>of</strong> B cell malignancies such as classical Hodgkin lymphoma<br />
(cHL). They discovered a number <strong>of</strong> molecular defects associ-<br />
<strong>of</strong>fenbar die Apoptose regulieren, indem sie den Transport von Cytochrom<br />

zeigen, dass mindestens drei neue Analoge hochwirksam (ED 50 zwischen<br />
0.01 und 0.05 µM) als selektive Inhibitoren der Replikation des<br />
B-Hepatitis-Virus sind. Dies rechtfertigt weitere Untersuchungen.<br />
Tumorimmunologie<br />
Homöostatische Chemokine und ihre Rezeptoren sind für die Migration<br />
von Lymphozyten zwischen und innerhalb von lymphoiden Geweben verantwortlich<br />
und deshalb notwendige Faktoren der Immunüberwachung<br />
und der Ausbildung von Autoimmuntoleranz. Die Gruppe von Martin<br />
Lipp ist speziell interessiert an der Rolle der homöostatischen Chemokine<br />
bei der Entstehung und Progression chronischer Entzündungen, die<br />
mit lymphoider Neogenese einhergehen, d.h. mit der Bildung lymphknotenähnlicher<br />
Gewebsstrukturen in der Umgebung von Entzündungsherden.<br />
Die Gruppe konnte beweisen, dass die Chemokin-Rezeptoren<br />
CXCR5 und CCR7 kritische Moleküle für die Signalübertragung bei der<br />
lymphoiden Neogenese bei chronischen Entzündungen sind. Für diese<br />
Studien wurde ein neuartiges Mausmodell für antigen-induzierte Arthritis<br />
(AIA) eingeführt, welches die effiziente Induktion ektopischer Lymphoidfollikel<br />
ermöglicht, einem Kennzeichen der rheumatoiden Arthritis<br />
beim Menschen. Ein weiterer Erfolg der Arbeitsgruppe war der Nachweis,<br />
dass CCR7 auch einen Schlüsselregulator der homöostatischen<br />
Lymphozyten-Rezirkulation durch Körperhöhlen und nicht-lymphoide<br />
periphere Gewebe darstellt.<br />
Autoreaktive T-Zellen, die den regulatorischen Mechanismen des<br />
Immunsystems entkommen sind, können schwere Autoimmunerkrankungen<br />
hervorrufen. Sie können jedoch auch den Schlüssel zu wirksamer<br />
Induktion tumorspezifischer Abwehrreaktionen sein, weil tumorassoziierte<br />
Antigene (TAA) ihrem Wesen nach Autoantigene sind, deren<br />
Wirkung den Bruch der Immuntoleranz erfordert. Die Arbeitsgruppe von<br />
Kirsten Falk und Olaf Rötzschke konnte als erste eine den<br />
Effektor/Memory T-Zellen ähnliche Subpopulation von T-Zellen beschreiben,<br />
die durch die Expression des Chemokinrezeptors CCR6 identifizierbar<br />
sind. Die Lokalisation und der Phänotyp dieser regulatorischen T REM -<br />
Zellen legt nahe, dass sie für die Kontrolle der potentiell schädlichen<br />
Autoimmunantworten im entzündeten Gewebe zuständig sind. Diese<br />
Befunde wurden unterstützt durch den Nachweis, dass Patienten mit<br />
Multipler Sklerose eine signifikant erniedrigte Anzahl solcher spezifischen<br />
T REM -Zellen aufweisen. Bemerkenswert bei dieser Subpopulation ist<br />
die Expression einer ecto-ATPase (CD39), was auf eine mögliche neuartige<br />
Funktion von extrazellulärem ATP in der Immunregulation hinweist.<br />
