amino acids by Cowan-Whittaker’s scale were derived by a direct chromatographic method
and due to this it was adapted for calculating ASH. The average hydrophobicity has given a
direct relationship with the retention of protein on the mentioned scales. This confirmed the
role of average hydrophobicity in the retention mechanism of HIC. The global averages of the
r 2 values were 0.85, 0.82 and 0.79 for the scales of Miyazawa-Jernigan, Cowan-Whittaker and
Tanford, respectively. The scales of the Cowan-Whittaker and Miyazawa-Jernigan have
revealed high correlation coefficient values than Tanford. It was also reported before that the
scales of Cowan-Whittaker and Miyazawa-Jernigan have provided the best correlation for the
prediction purposes in comparison to other scales (114).
Figure 15 A: Represents the average hydrophobicity calculated by the scale of Cowan-
Whittaker. The protein average hydrophobicity values of the three ligands were correlated with
the respective salt concentrations where the fractions were collected. The error bars represent
the standard deviations of the average hydrophobicity values in each fraction. The statistical
analysis revealed a significant correlation (r 2 = 0.82; p < 0.0001).
