probes hybridize and the two dyes come into close proximity. The energy emitted by<br />

the excited probe is transferred to the second probe, which emits red fluorescent light<br />

at a longer wavelength. At the polymerization step, both probes return into solution<br />

and the fluorescence signal returns to background.<br />

2.1.2.6. SCORPION PROBE<br />

Scorpion probe/primer systems combine sequence-specific primers and<br />

hybridization probes into a single structure, theoretically improving the kinetics<br />

of fluorescent signal generation (26,27). Fig. 9A shows the basic elements<br />

of a Scorpion probe. It consists of (1) a target-specific probe sequence held in<br />

a hairpin loop configuration by complementary 5� and 3� arms, (2) a
