10.3. DNA PURITY AND MARKER REPRODUCIBILTY<br />

The high purity of DNA is sine qua non to obtain reproducible results for any marker<br />

category. The common view is that some methods perceived as less sophisticated (RAPD<br />

or similar) do not need high purity of DNA and consequently many rapid protocols and commercial<br />

kits are used. On the other hand RAPDs have developed a reputation of poor reproducibility.<br />

As a result, having bad own experience, many reviewers reject papers based on<br />

RAPDs as unreliable. Conversely, own experience was that 100% of reproducibility can be<br />

obtained provided DNA is of high quality. Every time the isolation procedure was simplified,<br />

the results were much worse. Generally, commercial kits are not suitable for isolation. Instead
