SSAP polymorphism<br />

The primer complementary to L. perenne CACTA transposon, Tpo1 produced a great<br />

number of bands in L. multiflorum and L. perenne thus, confirming the high abundance of<br />

Tpo1 in these two ryegrasses. Similarly, each of the two LTR primers complementary to Ty1-<br />

copia consensus sequences produced bands on SSAP gel for each species, showing that<br />

each transposon is present within them (Figure 4.4A). Therefore, the transposons revealed<br />

by LTR1 and LTR2 were further assigned as Lolcopia1 and Lolcopia2, respectively.<br />

All transposon sequences studied were highly polymorphic among L. multiflorum and<br />

L. perenne (Table 4.1), indicating that they have been transposing within these species in<br />

the recent past. The overall polymorphism level for each transposon was the same for Tpo1<br />

and Lolcopia1 i.e., 93% and 88% for Lolcopia2. The proportion of polymorphic sites and the
