<str<strong>on</strong>g>Tecnai</str<strong>on</strong>g> <strong>on</strong>-<strong>line</strong> <strong>help</strong> <strong>Alignments</strong> 27<str<strong>on</strong>g>Tecnai</str<strong>on</strong>g> 12 Software versi<strong>on</strong> 26 Image HM-TEM Procedure6.1 Preparati<strong>on</strong> Image HM-TEM alignmentPurpose: Set up microscope for aligning the lower part of the column (the imaging system) in HM-TEM.Importance: ESSENTIAL to make sure that the alignment is d<strong>on</strong>e for the correct c<strong>on</strong>diti<strong>on</strong>s: centeredC2 aperture, eucentric height and specimen in focus.Method:C2 aperture centering:• Focus spot and center it <strong>on</strong> the screen• Turn INTENSITY overfocus (clockwise)• Center aperture until illuminated area is symmetrical around the screen center.Eucentric height: with the CompuStage switch <strong>on</strong> the Alpha wobbler and minimize image movement bychanging the Z height.6.2 Pivot point image shift HMPurpose: Align image shift pivot point = make sure that the diffracti<strong>on</strong> pattern does not shift when theimage shift is changed.Importance: ESSENTIAL for the proper functi<strong>on</strong>ing of image shifts.Method: Minimize the movement of the diffracti<strong>on</strong> pattern. The microscope 'wobbles' the image shiftwhich should cause no diffracti<strong>on</strong> shift.ProcedureThe alignment procedure c<strong>on</strong>sists of four steps:• Two preparati<strong>on</strong> steps for setting up the image and diffracti<strong>on</strong> pattern, respectively.• Two steps in which the X and Y pivot points are aligned.Note: Diffracti<strong>on</strong> focus. The Intensity setting is preset to a fixed value that gives a parallel incident beamwhich in turn should give a spot diffracti<strong>on</strong> pattern. If the pattern is not focused, it should be focused withthe diffracti<strong>on</strong> lens (FOCUS), not Intensity.Descripti<strong>on</strong>The alignment of the pivot points of the image deflecti<strong>on</strong> coils is essential to the proper functi<strong>on</strong>ing ofmany other alignments and calibrati<strong>on</strong>s: image shifts, diffracti<strong>on</strong> shifts, detector alignments andmeasurement functi<strong>on</strong>s.The image coils pivot points (image shift and diffracti<strong>on</strong> shift) alignment is reas<strong>on</strong>ably insensitive to theoperating c<strong>on</strong>diti<strong>on</strong>s and normally needs to be d<strong>on</strong>e <strong>on</strong>ly <strong>on</strong>ce. It is important that the image coils pivotpoints are aligned prior to aligning other parts of the microscope where the image coils are used,otherwise the latter alignments may have to be red<strong>on</strong>e. <strong>Alignments</strong> where the image coils are used arethose where images or diffracti<strong>on</strong> patterns are moved (image shift, diffracti<strong>on</strong> shift/diffracti<strong>on</strong> alignment).6.3 Pivot point diffracti<strong>on</strong> shift SAPurpose: Align the diffracti<strong>on</strong> shift pivot point = make sure the image does not move when the diffracti<strong>on</strong>shift is changed.Importance: ESSENTIAL for the proper functi<strong>on</strong>ing of image and diffracti<strong>on</strong> shifts.Method: Minimize the movement of the image.
