GENETIC ANALYSIS OF DNA POOLS 67genotyping on the genome scale to identify regions of thegenome associated with a disorder followed by pooledhaplotyping using single-molecule detection methods tonarrow down the associated regions. Once the associatedregions are mapped to 10-kb resolution, the regions canbe analyzed further by comprehensive DNA sequencingof individuals with the disorder and the controls. As thetechnological and theoretical methods improve, poolDNA analysis will be a powerful tool for genetic study ofcommon human disorders.ACKNOWLEDGMENTSThis work is funded by grants from the National Institutesof Health, the Smith Family Foundation, and theSandler Family Foundation. The authors thank US Genomics,Inc. and Perkin-Elmer Life Sciences Inc. for instrumentand reagent support.REFERENCESBarratt B.J., Payne F., Rance H.E., Nutland S., Todd J.A., andClayton D.G. 2002.
