Determination of Protein Structures in Solution by NMR

During the period 1979-83 methods were introduced for efficient sequence-specific
assignment of the 1H NMR spectra of proteins and nucleic acids in solution. This now provides
the basis for determination of the three dimensional structure of these biological macromolecules
(K. Wgthrich, NMR of Proteins and Nucleic Acids, Wiley, New York, 1986). This method for
structure determination will be surveyed, and novel approaches making use of isotope labeling and
X-filtering techniques will be described.
