Hemoglobin. (Hb). multiplicity. enables. fishes. to. adapt. themselves.with.different.ecological.and.envir<strong>on</strong>mental.c<strong>on</strong>diti<strong>on</strong>s..In.<br />
the. current. study,. the. Hbs. <strong>of</strong>. two. Iranian. Sturge<strong>on</strong>. species. have.<br />
been.investigated..After.extracti<strong>on</strong>.<strong>of</strong>.Hb.from.blood,.the.isoelectric.focusing.study.has.shown.that.both.species.have.multiple.Hbs.with.different.isoelectric.points.and.the.dominant.Hb.can.be.identified.from.other.Hb.comp<strong>on</strong>ents..The.dominant.Hbs.<strong>of</strong>.these.species.has.been.purified.using.I<strong>on</strong>-exchange.chromatography.<strong>on</strong>.CM-cellulose.<br />
column.. Fluorescence. spectrophotometery. studies. showed.<br />
that.the.Hbs.<strong>of</strong>.these.fishes.have.similar.structural.properties.with.<br />
each. other. and. clear. differences. with. human. Hb.. Comparing. the.<br />
heat. capacity. versus. temperature. <strong>of</strong>. different. Hbs. pr<strong>of</strong>iles. using.<br />
differential. scanning. calorimetry. (DSC). showed. varying. thermal.<br />
unfolding.characteristics..The.percentage.<strong>of</strong>.alpha-helix.and.betasheet.substructures.has.been.obtained.based.<strong>on</strong>.circular.dichroism.<br />
analysis.(CD).investigati<strong>on</strong>..UV–vis.spectrophotometer.was.used.<br />
to.measure.oxygen.affinity.using.sodium.dithi<strong>on</strong>ite..Oxygen.affinity.and.structural.properties.<strong>of</strong>.these.Hbs.were.compared.using.Hb.<br />
oxygen.dissociati<strong>on</strong>.curves..<br />

5’. Methylthioadenosine/S-adenosylhomocysteine. nucleosidase.(MTN).is.a.microbe.specific.enzyme.important.in.purine.and.<br />
methi<strong>on</strong>ine. salvage. from. byproducts. <strong>of</strong>. S-adenosylmethi<strong>on</strong>ine.<br />
dependent.reacti<strong>on</strong>s.such.as.polyamine.synthesis,.biological.methylati<strong>on</strong>,.<br />
autoinducer. and. vitamin. synthesis.. Interrupti<strong>on</strong>. <strong>of</strong>. MTN.<br />
activity.in.E. coli.leads.to.marked.decreases.in.intracellular.polyamine.c<strong>on</strong>tent.and.decreased.growth.rates.that.are.largely.reversible.<br />
by.polyamine.supplementati<strong>on</strong>..The.altered.polyamine.synthesis.is.<br />
most.likely.the.result.<strong>of</strong>.product.inhibiti<strong>on</strong>.<strong>of</strong>.polyamine.synthase.<br />
reacti<strong>on</strong>s. caused. by. the. accumulati<strong>on</strong>. <strong>of</strong>. 5’. Methylthioadenosine.<br />
(MTA).. Proteomic. analysis. also. reveals. that. decreased. levels. <strong>of</strong>.<br />
polyamine.synthases.accompany.MTN.gene.knock-out..Additi<strong>on</strong>al.<br />
effects.<strong>on</strong>.a.wide-range.<strong>of</strong>.metabolic.pathways.are.found.with.MTN.<br />
deficiency,.underscoring.a.complex.set.<strong>of</strong>.mechanisms.<strong>of</strong>.acti<strong>on</strong>.that.<br />
will.be.exerted.by.anti-MTN.specific.drugs,.and.suggesting.routes.<br />
<strong>of</strong>.drug.potentiati<strong>on</strong>.that.could.lead.to.potent.combinati<strong>on</strong>.therapies..<br />

2,3,7,8-Tetrachlorodibenzo-p-dioxin. (TCDD). is. an. envir<strong>on</strong>mental.<br />
pollutant. in. the. family. <strong>of</strong>. halogenated. aromatic. hydrocarb<strong>on</strong>s..<br />
Exposure. to. TCDD. elicits. a. spectrum. <strong>of</strong>. toxic. effects,.<br />
many.<strong>of</strong>.which.are.rooted.in.aberrant.cell.proliferati<strong>on</strong>,.activati<strong>on</strong>.<br />
and. differentiati<strong>on</strong>.. We. have. previously. shown. that. exposure. to.<br />
TCDD. suppresses. hepatocyte. proliferati<strong>on</strong>. in. a. mouse. model. <strong>of</strong>.<br />
liver. regenerati<strong>on</strong>. induced. by. 70%. partial. hepatectomy. (PH).. In.<br />
the.present. study,.we.tested.the.hypothesis.that.TCDD.alters.the.<br />
activati<strong>on</strong>.<strong>of</strong>. hepatic.stellate.cells.(HSCs),. which.regulate.regenerati<strong>on</strong>.<br />
through. the. producti<strong>on</strong>. <strong>of</strong>. soluble. mediators.. Mice. were.<br />
treated.with.TCDD.(20.µg/kg).or.peanut.oil.24.hr.prior.to.PH,.and.<br />
plasma.and.liver.tissue.were.collected.after.surgery..Exposure.to.<br />
TCDD.increased.expressi<strong>on</strong>.<strong>of</strong>.the.HSC.activati<strong>on</strong>.marker.alphasmooth.<br />
muscle. actin. and. increased. levels. <strong>of</strong>. m<strong>on</strong>ocyte. chemoattractant.<br />
protein. (MCP)-1,. a. chemokine. abundantly. produced. by.<br />
HSCs.. Likewise,. in. vitro. studies. using. a. human. hepatic. stellate.<br />
cell.line.revealed.that.TCDD.treatment.increased.proliferati<strong>on</strong>.and.<br />
MCP-1.producti<strong>on</strong>.and.impaired.the.ability.<strong>of</strong>.these.cells.to.store.<br />
retinoids�..Hence,.inappropriate.HSC.activati<strong>on</strong>.may.c<strong>on</strong>tribute.to.<br />

White. mature. adipocytes. have. been. shown. to. lose. their. fat.<br />
in.culture.and.de-differentiate.into.de-differentiated.(DFAT).cells..<br />
Immediately.after.the.loss.<strong>of</strong>.lipids,.DFAT.cells.express.several.stem.<br />
cell.markers.as.a.sign.<strong>of</strong>.multipotency..However,.it.is.not.known.for.<br />
how.many.cell.passages.the.cells.express.stem.cell.markers,.informati<strong>on</strong>.that.is.essential.in.defining.how.l<strong>on</strong>g.the.cells.can.be.used.<br />
a. source.<strong>of</strong>.multipotent.cells..To.test.the.l<strong>on</strong>gevity.<strong>of</strong>.DFAT.cell.<br />
expressi<strong>on</strong>.<strong>of</strong>.stem.markers,.mouse.DFAT.cells.were.cultured.for.up.<br />
to.10.passages..Cellular.RNA.was.extracted.after.each.passage,.and.<br />
cells.were.fixed.and.prepared.for.immunostaining.or.fluorescenceactivated.cell.sorting.(FACS)..Expressi<strong>on</strong>.was.determined.by.realtime.PCR..<br />
Real-time.PCR.showed.that.expressi<strong>on</strong>.<strong>of</strong>.Oct3/4,.Nanog.and.<br />
SOX2,.pluripotency.markers,.gradually.decreased.to.near.undetectable.levels.over.10.passages..After.~.2.passages,.25%.<strong>of</strong>.baseline.(passage.0).for.at.least.2.additi<strong>on</strong>al.passages..Optimally,.DFAT.cells.<br />
should.be.used.for.regenerative.purposes.no.later.than.two.passages.<br />
after.the.isolati<strong>on</strong>.<strong>of</strong>.adipocytes.<br />
