Partie C : Résultats – Chapitre C-IIIDISCUSSION1/ Evolution <strong>de</strong> la microflore bactérienne en présence <strong>de</strong> plomb et/ou <strong>de</strong> <strong>ver</strong>tel-00486649, <strong>ver</strong>sion 1 - 26 May 2010Les <strong>de</strong>nsités bactériennes cultivab<strong>les</strong> <strong><strong>de</strong>s</strong> agrégats racinaires quel que soit le niveau <strong>de</strong>contamination au plomb sont significativement plus élevées en présence <strong>de</strong> <strong>ver</strong>s. Les valeursobtenues entre 1,5 x 10 5 UFC/g <strong>de</strong> sol pour <strong>les</strong> échantillons AgP/T/SP et 1,1 x 10 8 UFC/g <strong><strong>de</strong>s</strong>ol pour <strong>les</strong> échantillons AgP/Pb++/SPV sont du même ordre <strong>de</strong> gran<strong>de</strong>ur que cel<strong>les</strong> décrites<strong>sur</strong> d’autres sites soit entre 10 5 et 10 8 UFC/g sol sec (Powlson et al., 2000). De nombreuxauteurs ont signalé que la présence <strong>de</strong> métaux <strong>lourds</strong> diminuait la biomasse microbienne soitdirectement soit en inhibant certaines propriétés biochimiques du sol indispensab<strong>les</strong> à leur<strong>sur</strong>vie (Kan<strong>de</strong>ler et al., 1996 ; Smit et al., 1997 ; Bååth et al., 1998 ; Konopka et al., 1999 ;Kuperman et Carreiro, 1997 ; Kelly et al., 1999 ; Ekelund et al., 2003). Cependant l’action<strong><strong>de</strong>s</strong> métaux est souvent spécifique, certains microorganismes résistants pouvant <strong>sur</strong>vivre à unstress environnemental, alors que d’autres plus sensib<strong>les</strong> disparaissent en présence <strong>de</strong> cesnouvel<strong>les</strong> contraintes (Giller et al., 1998). Ainsi, un certain nombre d’étu<strong><strong>de</strong>s</strong> n’ont pas mis enévi<strong>de</strong>nce la diminution <strong>de</strong> la biomasse microbienne en présence <strong>de</strong> métaux <strong>lourds</strong> (Knight etal., 1997 ; Landi et al., 2000 ; Fritze et al., 2000). Ceci résulterait du fait que parfois cettebaisse <strong>de</strong> biomasse est limitée dans le temps, elle peut en effet constituer une premièreréponse à un apport en métal et être ensuite suivie d’un retour à la valeur initiale à plus oumoins long terme (Kelly et al., 1999). Par ailleurs, <strong>les</strong> communautés bactériennes peuvent êtreinfluencées par <strong>de</strong> nombreux autres facteurs environnementaux tels que le pH, la température,la teneur en nutriments et la nature <strong><strong>de</strong>s</strong> exsudats racinaires… Ainsi, <strong><strong>de</strong>s</strong> variations dans labiomasse bactérienne ne sont pas systématiquement et directement imputab<strong>les</strong> à la présence<strong>de</strong> métal (Kelly et al., 1999 ; Bååth et al., 1998). Par exemple, si la contamination d’un solaffecte le développement <strong><strong>de</strong>s</strong> <strong>plante</strong>s, la réduction <strong>de</strong> la biomasse bactérienne observée n’estpas nécessairement due à un effet direct <strong>de</strong> la toxicité du métal <strong>sur</strong> <strong>les</strong> bactéries mais peut-êtreau manque <strong>de</strong> matière organique d’origine végétale (Konopka et al., 1999 ; Kuperman etCarreiro, 1997). La biomasse microbienne augmente en effet en présence <strong>de</strong> <strong>plante</strong>s, <strong>les</strong>exsudats racinaires servant <strong>de</strong> source <strong>de</strong> carbone et d’énergie pour <strong>les</strong> microorganismes du sol(Butenschoen et al., 2008). Dans notre cas, la pollution au plomb affecte favorablement ledéveloppement <strong>de</strong> la <strong>plante</strong> «phytoremédiatrice», ainsi que nous l’avons montré dans lepremier chapitre, on peut donc supposer que la présence <strong>de</strong> plomb en stimulant la croissance111
Partie C : Résultats – Chapitre C-IIIracinaire stimule également la production d’exsudats favorab<strong>les</strong> au développement <strong><strong>de</strong>s</strong>colonies microbiennes.tel-00486649, <strong>ver</strong>sion 1 - 26 May 2010Lorsque <strong>les</strong> <strong>plante</strong>s sont cultivées en présence <strong>de</strong> <strong>ver</strong>s <strong>de</strong> <strong>terre</strong>, dans le contexte <strong>de</strong> laphytoextration, l’action du <strong>ver</strong> provoque une augmentation significative <strong>de</strong> la <strong>de</strong>nsitébactérienne <strong><strong>de</strong>s</strong> agrégats racinaires <strong>de</strong> l’ordre <strong>de</strong> 20% aussi bien dans <strong>les</strong> traitements sansplomb que dans ceux à 500 mg Pb kg -1 ou 1000 mg Pb kg -1 . Ce résultat est très novateur car,si <strong>de</strong> nombreux travaux montrent que la <strong>de</strong>nsité et la di<strong>ver</strong>sité <strong><strong>de</strong>s</strong> micro-organismes sontmodifiées dans <strong>les</strong> agrégats et <strong>les</strong> galeries <strong>de</strong> <strong>ver</strong>s (Parle, 1963a ; Shaw et Pawluk, 1986 ;Scheu, 1987 ; Devliegher et Verstraete, 1997 ; Tiunov et Scheu, 1999 ; Tiunov etDobrovolskaya, 2002 ; Tiunov et Scheu, 2001 a et b), aucun travail ne s’était encore intéresséà l’influence <strong><strong>de</strong>s</strong> <strong>ver</strong>s <strong>de</strong> <strong>terre</strong> <strong>sur</strong> <strong>les</strong> communautés bactériennes <strong><strong>de</strong>s</strong> agrégats racinaires d’une<strong>plante</strong> « phytoremédiatrice ». Les <strong>interactions</strong> que <strong>les</strong> <strong>ver</strong>s entretiennent avec <strong>les</strong> microorganismesont cependant probablement joué un rôle essentiel dans notre étu<strong>de</strong>.2/ Effet du Pb et/ou du <strong>ver</strong> <strong>sur</strong> la structure <strong><strong>de</strong>s</strong> communautés bactériennesD’après <strong>les</strong> résultats obtenus en PCR-DGGE, la pollution <strong><strong>de</strong>s</strong> sols par du plomb a unimpact <strong>sur</strong> ces communautés bactériennes, le changement <strong>de</strong> structure étant plus net lorsque leniveau <strong>de</strong> pollution est le plus élevé. Hu et al. (2007) ont déjà montré qu’il y avait <strong><strong>de</strong>s</strong>différences significatives <strong>de</strong> la structure <strong><strong>de</strong>s</strong> communautés microbiennes dépendant du niveau<strong>de</strong> contamination en plomb.Par contre, il apparaît que la structure <strong><strong>de</strong>s</strong> communautés bactériennes présentes dans <strong>les</strong>agrégats racinaires n’est pas modifiée par <strong>les</strong> activités <strong><strong>de</strong>s</strong> <strong>ver</strong>s P. corethrurus alors que la<strong>de</strong>nsité et l’activité phytoremédiatrice <strong>de</strong> la <strong>plante</strong> le sont fortement. Plusieurs hypothèsespeuvent être formulées pour expliquer ce résultat <strong>sur</strong>prenant :‣ Tout d’abord, <strong>les</strong> techniques <strong>de</strong> « fingerprint » moléculaire (ou empreintesgénétiques) ciblant <strong>les</strong> aci<strong><strong>de</strong>s</strong> nucléoti<strong><strong>de</strong>s</strong> comme <strong>les</strong> techniques DGGE, TGGE,dépen<strong>de</strong>nt <strong>de</strong> l’extraction <strong><strong>de</strong>s</strong> aci<strong><strong>de</strong>s</strong> nucléi<strong><strong>de</strong>s</strong> <strong><strong>de</strong>s</strong> sols, du choix <strong><strong>de</strong>s</strong> amorces et <strong>de</strong> leuramplification par PCR ; el<strong>les</strong> sont donc soumises aux biais et limites associés à cesétapes clés (Martin-Laurent et al., 2001 ; Robe et al., 2003 ; Wintzingero<strong>de</strong> et al.,1997 ; Hongoh et al., 2003) :112
