ToWARdS The uNdeRSTANdINg of geNeTIC ANd moLeCuLAR BASIS of heTeRoSIS IN WheAT (TRITICum AeSTIVum L.) Sun Qixin, Ni Zhongfu, Peng Huiru, Yao Yingyin, Du Jinkun, Liu Gang, Lu Lahu Department of Plant Genetics and Breeding, China Agricultural University, Beijing, 100193, China E-mail Address of presenting author: qxsun@cau.edu.cn In China, more than 10 hybrid wheat cultivars have been registered by 2007, and hybrid wheat is planted in more than 10000 hectares annually, with yield advantage of 20%. Despite the extensive efforts in hybrid wheat breeding, however, mechanisms of heterosis are largely unknown. Attempts have been made in our research group for last decade to understand genetic and molecular basis of heterosis in wheat: 1) An immortalized F2 (IF2) population derived from the cross between common wheat (T. aestivum L.) 3338 and spelt wheat (T.spelta L.) Altgold was used to investigate the genetic basis of heterosis. A total of 39 heterotic loci (HL) were detected, among which 23 showed negative dominance effect, which resulted in less net dominance effect of these traits. In addition, 27 showed over dominance effect, suggesting that over dominance effect at single locus is the main genetic basis of heterosis in the hybrid; 2) Comparative transcriptomic and proteomic profiles between hybrids and their parental inbreds were performed, and all possible modes of gene action, including additivity, high- and low-parent dominance, underdominance, and overdominance are detected, which may be responsible for the observed heterosis; 3) Differential gene expression patterns in leaves between hybrids and their parental inbreds with heterosis in a wheat diallel cross were investigated. The results demonstrated that some patterns of differential expression detected in leaves at jointing stage and heading stage are correlated with heterosis in some traits, and heterosis in some other traits are correlated with differential expression patterns only detected in leaves of either of the developmental stages; 4) Differentially expressed gene/proteins involving in diverse of functional categories were identified and the their functions were also analyzed. Most notably, genes that were upregulated in wheat hybrids are positively correlated with trait performance, but negatively for downregulated, which provide transgenic evidence for the notion that changes in gene expression between hybrids and their parents is responsible for the heterosis; 5) Two EST-SSR markers were selected for allelespecific expression analysis in wheat, and unequal expression of the two alleles (biallelic) in uppermost internode of heading stage was detected in a group of cross combinations. Importantly, allelic expression variation is positively correlated with plant height of some specific hybrids; 6) Heterosis in internode elongation and plant height are commonly observed in wheat hybrid plants. Our analysis revealed that higher GA contents in hybrid plants could result from the differential expression of genes that participate in the regulation of GA biosynthesis and catabolism, and increased stem growth rate of hybrids could be related to the differential expression of genes participating in regulation of GA 340
esponse pathways. Collectively, a model for GA biosynthesis and response pathway in regulation of heterosis in plant height of wheat was proposed.
