Sensitivity<br />

Clinitest reagent tablets will detect as little as 250mg of sugar in 100ml of<br />

urine.<br />

Specific ( Enzymatic ) Tests<br />

Enzymatic tests are specific tests for glucose. <strong>The</strong>y are reagent strips<br />

(dipsticks ), which are impregnated with enzymes glucose oxidases.<br />

Glucose oxidase catalyzes only the oxidation glucose to gluconic acid<br />

and hydrogen peroxide. <strong>The</strong> principle of all enzymatic, which is based<br />

on the uses of glucose oxidase, is the same. <strong>The</strong>y differ only on the<br />

uses of different type of chromogen (a color indicator ).<br />

A. Clinistix Reagent Strip Test<br />

Principle<br />

This is a specific test for glucose based on the use of the enzyme<br />

glucose oxidase, which is impregnated on a dip strip. In this test glucose<br />

oxidase oxidizes glucose to gluconic acid and at the same time<br />

reduces atmospheric oxygen to hydrogen peroxide. <strong>The</strong> hydrogen<br />

peroxide formed , in the presence of the enzyme peroxidase,<br />

oxidizes the reduced form of o-toluidine( a chromogen ) to oxidized<br />

form of the indicator, wich produces a color change proportional to<br />

the amount of glucose in the urine. Other sugars are not substrates<br />

for the enzyme do not react with this test.<br />

A positive reaction is seen as a change of color from red to blue,<br />

depending on the amount of glucose present in the urine.
