The Marie Curie project Pipkigko has made progress on therapies for this andother developmental disorders such as spina bifida, bleeding maladies and even certain forms of cancer.
When cells don’tstick togetherDr Kyle Legate, a Canadian scientist with a PhD in biochemistry,was lured to Europe by means of an Incoming InternationalFellowship under the Marie Curie programme. While here, hemade signifi cant strides towards understanding the mechanismand consequences of cell adhesion using the mouse as a modelfor the biological system. He spent his two-year fellowship atDr Reinhard Fässler’s laboratory in the Department of MolecularMedicine at the Max Planck Institute of Biochemistry, Martinsriedin Germany.‘Cell adhesion’, the focus of Pipkigko, can be defi ned as thebinding of a cell to a surface, extracellular matrix or anothercell using cell-adhesion molecules. Put simply, it involves cellscoming together and bonding.The Pipkigko project examined the function of a particularenzyme that is present in specialised cell-adhesion structurescalled ‘focal adhesions’. These adhesions are specifi c typesof large macromolecular assemblies through which bothmechanical force and regulatory signals are transmitted. Theenzyme Dr Legate examined is known as phosphatidylinositol4-phosphate 5 kinase Type I gamma (PIPKIγ). It makes amembrane lipid (the cell membrane is a fl at sheet that forms acontinuous barrier around a cell), called PIP2, of high importancefor many cell functions, including cell adhesion.Location, location, location‘PIPKIγ is of particular interest because its location within thecell makes it well placed to regulate the functions of many celladhesionproteins. My work involved examining the consequencesof disturbing the location of PIPKIγ within the cell so that it isunable to perform its function,’ says Dr Legate. ‘The researchshows that mice develop normally when the localisation ofPIPKIγ to focal adhesions is disrupted. However, cells derivedfrom these mice are signifi cantly affected,’ he continues.Several parameters linked to adhesion are sluggish in cellslacking PIPKIγ, compared to cells in which the enzyme islocalised normally.
