Restauration of SHIP-1 activity in human leukemic cells modifies NF-!B activation pathway and cellular survival upon oxidative stress treatment Geoffrey Gloire*, Edith Charlier*, Souad Rhamouni*, Cédric Volanti*, Alain Chariot*, Christophe Erneux $ and Jacques Piette* *University of Liège, B-4000 Liège, $ University of Brussels, B-1070 Brussels Since the discovery that NF-!B 1 could be activated by H 2O 2, several laboratories put a lot of efforts into dissecting the molecular mechanisms underlying this activation, particularly in Jurkat leukemic cells. These works led to the observation that NF-!B activation by H 2O 2 involved tyrosine phosphorylation of the inhibitor I!B" through an IKK-independent mechanism. In the present work, we investigated with more details NF-!B redox regulation in human leukemic cells. By using different cell lines (CEM, Jurkat and Jurkat JR), we clearly showed that NF-!B activation by H 2O 2 is cell-type dependent: it activates NF-!B through tyrosine phosphorylation of I!B" in Jurkat cells, but induces an IKK-mediated IkB" phosphorylation on serines 32 and 36 in CEM and Jurkat JR cells. We showed that this H 2O 2induced IKK activation in CEM and Jurkat JR cells is mediated by SHIP-1, a lipid phosphatase which is absent in Jurkat cells. Indeed, the genetic complementation of SHIP-1 in Jurkat cells made them shifting to an IKK-dependent mechanism upon oxidative stress stimulation. The phosphatase domain of SHIP-1 is crucial in that process, since the complementation of Jurkat cells with a mutant of SHIP-1 lacking catalytic activity failed to mediate IKK activation upon H 2O 2 treatment. We also showed that Jurkat expressing SHIP-1 are much more resistant to H 2O 2-induced apoptosis than the parental cells, suggesting that SHIP-1 have an important role in protecting leukemic cells from ROS-induced apoptosis. Overall, our study highlight a novel role for SHIP-1 in leukemic cells ROS responses in terms of signal transduction pathways and apoptosis resistance, which can be of interest in improving ROS-mediated chemotherapies. - 81 -
